雌激素受体相关受体α与雌激素受体在子宫内膜癌组织中的表达

摘　　要：目的雌激素受体(estrogen receptor,ER)α、β在子宫内膜癌的发生发展中起着非常重要的作用。有研究提示,雌激素受体相关受体α(estrogen receptor-related receptorα,ERRα)与ER竞争配体也参与子宫内膜癌的发生,但详细的机制尚不明了。文中研究的目的是检测ERRα和ERα、ERβ在子宫内膜癌中mRNA的表达水平,并探讨其关系。方法采用real-time TaqMan PCR定量的方法,检测37例子宫内膜癌组织(内膜癌组)、25例癌旁组织(癌旁组)以及20例正常子宫内膜(对照组)中ERRα和ER亚型mRNA的表达量。结果3组患者中所有受体均有表达,在内膜癌组中,ERα和ERβ表达水平较对照组明显下降(P<0.05),ERRα的表达水平明显升高(P<0.05)。癌旁组患者的所有指标与癌组织和正常内膜组织相比均显著升高(P<0.05)。结论子宫内膜癌组织中ERα、ERβ低表达,而ERRα相对高表达,癌旁组织中3种受体均为高表达,说明癌组织中部分ERα、ERβ失活,ERRα在内膜细胞向癌细胞进展过程中起着非常重要的调节作用。Objective Estrogen receptors α （ERα） and β （ERβ） play a vital role in the development of endometrial cancer. Estrogen receptor-related receptor α （ERRα） reportedly competes with estrogen receptors for the same legend and participates in the genesis of endometrial cancer, but its mechanism remains unclear. The aim of this study was to analyze the mRNA expressions of ERRα, ERα and ERβ in endometrial carcinoma. Methods We detected the mRNA expressions of ERRα, ERα and ERβ in 37 cases of endometrial carcinoma and 25 samples of paracancer by real time Taqman PCR. Another 20 normal samples served as a control group. Results Compared with the control group, the expressions of ERa and Erβ were significantly decreased （P 〈 0.05）, while that of ERRα remarkably increased in the malignant endometrial specimens （ P 〈 0.05 ）. RT-PCR results showed that the expressions of all the receptors were significantly higher in the paracancer samples than in the other two groups （ P 〈 0.05 ）. Conclusion The expressions of ERα and ERβ were relatively low, and that of ERRα high in endometrial carcinoma, while all the three receptors were highly expressed in paracancer tissues. ERRor plays a crucial role in the development of endometrial carcinoma.
