摘 要:目的:探讨蟾毒灵对人肝癌细胞HepG2细胞周期的影响及其相关机制。方法:MTT方法检测蟾毒灵单体对肝癌细胞HepG2的生长抑制作用;FCM法检测蟾毒灵单体诱导HepG2细胞周期阻滞的作用;Western blot方法观察蟾毒灵单体对细胞周期相关蛋白的作用。结果:蟾毒灵能够抑制HepG2细胞的生长,该抑制作用呈时间和剂量依赖性。蟾毒灵能够诱导HepG2细胞周期阻滞于G2/M期。蟾毒灵对HepG2细胞周期的阻滞作用可能与cyclinB1、CDK1的表达下调有关。结论:蟾毒灵能够通过下调细胞周期相关蛋白的表达,诱导HepG2细胞阻滞于G2/M期。Objective:To study the effect of bufalin on hepatocellular carcinoma cell line HepG2 and the involved mechanism. Methods:Inhibitory effect of bufalin on the proliferation of HepG2 cells was determined by MTT assay; cell cycle distribution was shown by flow FCM ; Western blot was performed to detect the expression of protein related to cell cycle. Results:Bufalin could inhibit the growth of HepG2 cell in a concentration and time dependent manner in vitro. Bufalin could cause G2/M cell cycle arrest in HepG2 cell. The effect on cell cycle was partly due to the decrease of cyclinB1 and CDK1. Conclusion : Bufalin could induce G2/M arrest of HepG2 cell by dowrtregulafiing the expres- sion of critical molecules related to cell cycle.
