摘　　要：目的:建立H460细胞三维立体培养模型,探讨VEGF对H460细胞、MMP-2表达的影响。方法:采用非小细胞肺癌(NCLC)H460细胞系建立肺癌细胞三维立体培养模型,随机分成3组,给与不同浓度的VEGF,明胶酶谱法测定3组MMP-2的表达情况。结果:各组H460细胞生长良好;各实验组的H460细胞在不同浓度的VEGF和不同时间的作用下,细胞内的MMP-2的表达有差异。结论:VEGF能上调H460细胞MMP-2表达,且促进作用具有剂量-时间依赖性;三维立体培养模型有利于观察和研究基质金属蛋白酶在肿瘤侵袭和转移的作用。Objective ： To evaluate the stimulus of VEGF on MMP - 2 expression of H460 cells in three - dimen- sional culture collagen gels. Methods ： H460 cells were cultured in three - dimensional culture collagen gels, different does of VEGF was given to different groups. The expression of MMP -2 was detected by gelatin zymography. Results： 14460 cells grow well in three - dimensional culture collagen gels. The expression of MMP - 2 was different in different VEGF concentration and at different time. Conclusion： The activity and expression of MMP - 2 were enhanced by VEDF, and the stimulus is does - time dependent ; the three - dimensional culture collagen gels provided a reliable re- search tool for studying the stimulus of MMPs in the invasion and metastasis of cancer.
