摘　　要：通过第１天在外周血淋巴细胞中加入γ－ＩＦＮ，第２天再加入ＩＬ－２、ＣＤ３单抗和ＩＬ－１，获得了已被定义为多种细胞因子诱导的杀伤细胞（Ｃｙｔｏｋｉｎｅｉｎｄｕｃｅｄｋｉｌｅｒｃｅｌｓ）即ＣＩＫ细胞。通过这种方法，使外周血中微量的ＣＤ３＋ＣＤ５６＋细胞得到大量扩增。这种ＣＤ３＋ＣＤ５６＋细胞被证明是Ｔ细胞并具有ＮＫ细胞表面标志ＣＤ５６抗原。ＣＩＫ能溶解多种肿瘤细胞，表现为非ＭＨＣ限制性杀伤，杀伤活性远高于ＬＡＫ细胞。The authors report a novel protocol for generation of highly efficient cytotoxic effctor cells which have been termed cytokine induced killer(CIK) cells by culturing PBMC in the presence of γ IFN on day 1,and IL 2,mAb against CD3,IL 1 on day 2.Using this protocol,large numbers of double positive CD3 +CD56 + cells can be generated,which are rare in uncultured PBMC.The CD3 +CD56 + cells are proved to be T cells co expressting CD56 antigen.CIK cells have been found to be capable of killing numberous tumor cells,and the cytotoxic activity is not MHC restricted and more efficient than that of LAK cells.
