摘  要:目的观察三氧化二砷(As2O3)对人胆管癌细胞QBC-939黏附、侵袭力的影响,并探讨其机制。方法常规培养QBC-939,之后加入As2O3培养。MTT法检测细胞黏附力,Transwell趋化小室检测细胞侵袭力,RT-PCR检测细胞的尿激酶型纤溶酶原激活因子(u-PA)mRNA、基质金属蛋白酶-9(MMP-9)mRNA。结果加入As2O3培养后,QBC-939细胞黏附力、穿膜细胞数较对照组明显下降(P均<0.05),其u-PA、MMP-9 mRNA表达减少(P均<0.05),并呈时间、剂量依赖性。结论As2O3可抑制QBC-939的黏附和侵袭力,其机制与降低该细胞u-PA、MMP-9 mRNA表达有关。Objective To observe the effect of arsenic trioxide ( As2O3 ) on adhesion and invasiveness of humoal cholangiocareinoma eeUs line QBC-939 and investigate its mechanism. Methods Cholanglocareinoma ceils ( QBC-939 ) were cultured regularly and treated by As2O3. Adhesion of QBC-939 was assessed by MTr,invasiveness of QBC-939 was detected by Transwel/chamber,the u-PA mRNA and MMP-9 mRNA in QBC-939 were detected by RT-PCR. Results After QBC-939 were treated with As2O3, the adhesion and invasiveness of QBC-939 was signifieandy decreased compared with the control group( P 〈 0.05 ) , the expression of u-PA ,MMP-9 mRNA were decreased( P 〈 0.05 ) in time- and dose-dependent manner. Condusion As2O3 can inhibit the adhesion and invasiveness of QBC-939,the mechanism may be reducing the expression of u-PA and MMP-9 mRNA.
