含氮二磷酸盐联合IL-2对肾癌患者外周血γδT细胞增殖的影响

摘　　要：目的探讨含氮二磷酸盐联合IL-2(N-BPS/IL-2)对肾癌患者外周血γδT细胞体内诱导增殖的作用。方法①采用流式细胞仪分别测定试验组26例未经任何治疗的肾癌患者和对照组42例健康人外周血总T细胞绝对值、γδT细胞绝对值、γδT细胞相对值。②测定26例肾癌患者手术治疗后外周血总T细胞绝对值、γδT细胞绝对值、γδT细胞相对值。③26例肾癌患者行N-BPS/IL-2体内诱导γδT细胞增殖的免疫治疗,测定治疗前后外周血总T细胞绝对值、γδT细胞绝对值、γδT细胞相对值,与对照组比较。结果①肾癌患者和健康人群外周血总T细胞绝对值比较,差异无统计学意义(P>0.05);γδT细胞绝对值、γδT细胞相对值分别比较,低于健康人群水平,差异有统计学意义(P<0.001)。②26例肾癌患者手术治疗前后外周血总T细胞绝对值、γδT细胞绝对值、γδT细胞相对值比较,差异无统计学意义(P>0.05)。③26例肾癌患者N-BPS/IL-2体内诱导γδT细胞增殖治疗后外周血总T细胞绝对值、γδT细胞绝对值、γδT细胞相对值显著高于治疗前,差异有统计学意义(P<0.01);与对照组健康人群比较,差异无统计学意义(P>0.05)。结论肾癌患者外周血γδT细胞计数显著低于健康人群;手术对肾癌患者外周血γδT细胞计数无明显影响;N-BPS/IL-2在体内可诱导肾癌患者外周血γδT细胞显著增殖。Objective To investigate the proliferation function of gamma delta T （γδT） cells in peripheral blood by IL-2 and nitrogen containing-bisphosphonates （N-BPS） -stimulated on patients with renal carcinoma.Methods T cells absolute value,γδT cells absolute value and γδT cells relative magnitude in peripheral blood were determined by γδT cells proliferation assay in 26 patients with renal carcinoma without any treatment and 42 healthy individuals by flow cytometry.T cells absolute value,γδT cells absolute value and γδT cells relative magnitude before and after operation were determined among 26 patients in trial group.In trial group,immunotherapy of γδT cells proliferation by IL-2 and N-BPS -stimulated in vivo was used.Then T cells absolute value,γδT cells absolute value and γδT cells relative magnitude in peripheral blood before and after the immunotherapy were determined,taking the 42 healthy individuals as control group.Results Difference was not significant in the absolute count of all T cells between patients with renal carcinoma and healthy individuals （P 〉 0.05）.γδT cells absolute value and the percent of γδT cells in patients with renal carcinoma were lower than those in healthy individuals.There were significant differences between those indexes in two groups （P 〉 0.001）.Differences were not significant between the T cells absolute value,γδT cells absolute value and γδT cells relative magnitude in peripheral blood of the 26 patients before operation and those indexes after operation （P 〉 0.05）.In trial group,the T cells absolute value,γδT cells absolute value and γδT cells relative magnitude in peripheral blood increased after the immunotherapy of γδT cells proliferation by IL-2 and N-BPS -stimulated in vivo.There were significant differences between those indexes before and after treatment （P 〈 0.01）.While there were no significant differences in those values between experimental group and control group （P 〉 0.05）.Conclusions γδT cells in
