摘  要:目的:探讨通络益智散对血管性痴呆大鼠海马CA1区P53蛋白表达的影响。方法:用双侧颈总动脉(CCA)法制备大鼠血管性痴呆模型,将大鼠随机分为假手术组、模型组、通络益智散高低剂量组和尼莫地平组。3周后,断头处死大鼠,取大脑海马做病理和海马CA1区P53免疫组化检测。结果:假手术组海马CA1区未见P53阳性细胞,通络益智散大小剂量及尼莫地平组海马CA1区P53阳性细胞平均灰度值均明显高于模型组(P<0.05)。结论:通络益智散能显著降低CA1区P53蛋白的表达,抑制血管性痴呆大鼠脑损伤引起的细胞凋亡,对血管性痴呆大鼠的脑组织有保护作用,这可能是其治疗血管性痴呆的作用机理之一。Objective: To explore the effect of Tongluo Yizhi power (TLYZP)on expression of P53 protein in hippoeampus CA1 area of rat.with vascular dementia. Methods: The rat model of vascular dementia was build by repeated legation of bilateral common carotid arteries (CCA)together. The rats were randomly divided into sham operation group, model group, TLYZP high and low dose group, Nimodipine group with 10 rats in each group. After 3 weeks, the expression of P53 in hippocampus CA1 were detected. Results : The sham-operated group of hippocampus CA1 area has no P53-positive cells. The average gray value of P53 positive cells of hippocampus CA1 area in TLYZP and nimodipine group were significantly higher than the model group (P〈0.05). Conclusion: TLYZP could prominently decrease the expression of P53 and inhibit apoptosis of va.seular dementia that caused by brain injury in rats. It has a protective effect on brain tissue, which could be one of mechanism for treatment vascular dementia.
