摘　　要：目的:探讨蛋白激酶B(Protein kinase B,PKB/Akt)在肝细胞肝癌(Hepatocellular carcinoma,HCC)组织及癌旁肝组织(Liver tissue just around the tumor,LAT)中的表达变化,并分析其与HCC临床分级之间的关系。方法:采用逆转录聚合酶链反应(Reverse transcription-polymerase chain reaction,RT-PCR)、免疫印迹(Western blot)以及免疫组织化学染色方法检测HCC组织标本中癌及癌旁Akt的表达变化,并分析Akt的表达与HCC临床分级之间的关系。结果:Akt在HCC癌组织中的表达高于癌旁组织,Akt在癌组织中表达的阳性强度与临床分级呈正相关,癌组织中过表达的Akt在细胞核的定位明显增强。结论:Akt的过表达在HCC的发生、发展中具有重要作用。Objective： To investigate the expression of Akt proteins in hepatocellular carcinoma （HCC） and it＇s relation with clinical grading. Methods： Firstly, the mRNA levels of Akt in HCC and liver tissue just around the tumor（LAT） were detected by semi-quantitative reverse transcription and the polymerase chain reaction （RT-PCR）. Then, analyzing the expression of Akt proteins in HCC and LAT by Western blot and immunohistochemical methods. Finally, the relation between Akt proteins level and HCC clinical grading was analyzed. Results： The expression of Akt in HCC are much higher than LAT, and there is a positive correlation between Akt proteins level and HCC clinical grading. Furthermore,Akt is highly concentrated in the nucleus of HCC. Conclusion： The over-expression of Akt may play an important role in the occurrence and development of HCC.
