摘  要:目的:探讨超声定位辐照载紫杉醇脂质微泡(Paclitaxel-carrying liposome microbubbles,PLM)的方法对裸鼠人卵巢癌移植瘤的抑瘤效应及可能的机制。方法:建立20只人卵巢癌裸鼠皮下移植瘤模型,随机分成PLM辐照组、紫杉醇药物组、PLM组及生理盐水对照组,每组5只,分别给予不同处理,末次处理24 h后处死裸鼠,测量重量体积,计算抑瘤率;用免疫组织化学SP法检测肿瘤组织中细胞核相关抗原Ki-67并进行半定量分析;在透射电镜下对肿瘤细胞进行形态学观察。结果:与对照组比较,PLM辐照组及紫杉醇药物组对移植瘤的生长有明显的抑制作用,以PLM辐照组的抑瘤率最高(P<0.01);PLM辐照组及紫杉醇药物组肿瘤组织中Ki-67的阳性表达量均下降,前者下降更为明显(P<0.01)。透射电镜下观察到PLM辐照组肿瘤凋亡细胞出现较多。结论:与紫杉醇药物组相比,超声破坏PLM的方法对裸鼠SKOV3卵巢癌皮下移植瘤具有较强的体内抑瘤效应,可能是通过诱导细胞的凋亡,抑制肿瘤细胞的增殖活性而延缓肿瘤生长的。Objective:To discuss the inhibitory effect of ultrasound fitted irrad-iation paclitaxel-carrying liposome microbubbles (PLM) agent on nude micetran-splanted human ovarian carcinoma and its possible mechanism of action. Methods: We established nude mice transplanted SKOV3 human ovarian carcinoma. Then mice were randomly divided into 4 groups (5 mice in each group). Each group was treated in a different way.The rate of tumor inhibition was calculatedafter sacrificing the animals at the end of experiment.The nuclear associated an-tigen Ki-67 was measured immunohistochemieally and analyzed through compnt-er.Tumor tissues were observed under the transmission electrion microscope (TEM). Results: Compared with control, the growth of tumor in PLM combined ultr-asound group and paclixtaxel chemotherapy group was slow and the inhibitory effect of tumor was significant (P〈0.01).The Ki67 expression were all decreased(P〈0.01 ).But the former which inhibitory effect of tumor was much more signi-fieant (P〈0.01).Apoptosisbodies were observed under transmission electron micr-oscope and the PLM combined ultrasound group was stronger than other groups.Conclusion:Ultrasound destructing PLM agent has good therapeutic effect in the treatment of nude mice transplanted human ovarian carcinoma.The mechanism of action may be its antiangiogenetic effect, interrupting, inhibiting proliferation and inducing apoptosis.
