摘  要:本文首次对厚壳树叶75%乙醇粗提物及不同萃取部位进行DPPH(1,1-二苯基苦基苯肼),还原能力及亚铁离子螯合力的抗氧化活性研究,DPPH实验的IC50值(μg/mL)分别为:总提取物(18.554-0.32μg/mL)、V。(21.09±0.25μg/mL)、乙酸乙酯层(31.694-0.61μg,/mL)、二氯甲烷层(39.06±1.61μg/mL)、水层(69.184-9.40μg/mL)、正丁醇层(81.64±3.18μg/mL);还原能力由强到弱分别为Vc〉乙酸乙酯层〉总提取物〉正丁醇层〉水层〉二氯甲烷层,在样品浓度50la,g/mL时亚铁离子螯合力测试抑制率由强到弱分别为乙酸乙酯层(38.21%),正丁醇层(34.31%)、Vc(34.17%)、水层(32.95%)、总提取物(28.83%)、二氯甲烷层(31.30%);结果表明乙酸乙酯部位具有相对较强的抗氧化活性(IC50);故首次建立了HPLC/UV/MS法对抗氧化活性乙酸乙酯部位的多酚类成分进行了快速鉴别,为该种植物的进一步应用提供科学依据。The antioxidant activities of Ehretia thyrsiflora leave crude extract and the four fractions of dichloromethane, ethyl acetate, n-butanol, and water were tested using different in vitro assays of the scavenging effects on DPPH ( 1,1-di- phenyl-2-picrylhydrazyl) ,reducing power and Fe2+ -chelating activity. The IC50 values(p.g/mL) in DPPH test were CE ( 18.55 ±0.32 μg/mL) ,ASe(21.09 ±0.25 μg/mL) ,EaF(31.69 ±61 μg/mL) ,CfF(39.06 ±1.61 μg/mL) ,WtF (69.18 +9.40 μg/mL) ,nBF(81.64 ±3.18 μg/mL) ,respectively. The EaF had better reductive effect than all the other fractions with the orders ASe 〉 EaF 〉 CE 〉 nBF 〉 WtF 〉 CfF and the best inhibitory effect with 38.21% of chela- ring ability at a concentration of 50 I^g/mL, followed by nBF ( 34. 31% ), ASc ( 34. 17% ), WtF ( 32. 95% ), CE (28.83%), CfF (31.30%), respectively. The ethyl acetate fraction which showed stronger antioxidant potentials was chemically profiled using high performance liquid chromatography with electrospray ionization-mass spectrometric detector(HPLC/UV/ESI-MSD). The separated compounds were individually analyzed by their molecular ions, characteristic fragment ion peaks and UV data,and in comparison with the standard. A total of 13 polyphenols were identified.
