摘 要:利用SAS,Minitab和Design-Expert软件,采用Placket-Burman设计和中心组合设计对盐霉素的发酵条件进行优化。并运用响应面方法对盐霉素发酵条件进行进一步的分析,根据建立的多元二次方程确定其最佳水平,得到最佳的培养基:糊精4.5%,玉米浆0.6%,黄豆粉0.3%,氯化钾0.3%,硝酸铵0.3%,磷酸氢二钾0.04%,豆油10%,碳酸钙0.1%。培养条件为:接种量10%,pH7.0,摇床转速为250r/min,培养周期10d。在此最优化条件下得到盐霉素发酵单位比优化前的产量提高了44%。The fermentation condition of salinomycin prodction was optimized through Plackeet-Burman experimental design and central composite design (CCD) with the aid of SAS, Minitab and Design-Expert soflwares. The levels of fermentation condition were further screened and identified by response surface methodolgy. From the resulting of multi-quadratic equation, the optimized levels of influencing factors could be estimated. The optimized medium was obtained as follows: dextrin 4.5%, corn steep liquor 0.6%, soybeen meal 0.3%, KCl 0.3%, NH4NO3 0.3%, KH2PO4 0.04%, oil 10%, CaCO3 0.1% and initial pH 7.0. The optimized culture conditions were also established: shaker speed 250r/min, inoculum size 10% V/V, and cultivation duration for 10 days. The salinomycin production titer under this developed conditions in shaking flask culture was increased by 44% compared with that obtained at the unoptimized conditions, indicating the newly developed fermentation conditions for salinomycin production has a promising application in the industrial production.
