肝内趋化因子CCL21和CCL25在不同肝组织中的表达变化

摘　　要：目的观察正常肝、肝纤维化和肝硬化患者肝组织内趋化因子CCL21和CCL25的表达变化。方法ELISA方法检测9例正常肝、10例肝纤维化、11例肝硬化患者肝组织中CCL21和CCL25的含量。结果正常组、肝纤维化组、肝硬化组的CCL21含量分别为(0.14±0.21)、(0.92±0.69)和(3.22±1.45)pg/mg;肝硬化组的含量高于正常组和肝纤维化组,肝纤维化组和正常组CCL21的含量差异无统计学意义。正常组、肝纤维化组、肝硬化组的CCL25含量分别为(4.06±2.23)、(14.46±4.10)和(21.97±2.98)pg/mg;肝硬化组的含量高于正常组和肝纤维化组,肝纤维化组的含量高于正常组。结论肝脏发生纤维增生性损伤时,肝内CCL21和CCL25的含量显著增加,随着病变由肝纤维化向肝硬化阶段发展,肝内CCL21和CCL25含量也进一步升高,提示CCL21和CCL25很可能是肝纤维化和肝硬化状态下趋化骨髓源性肝干细胞迁移到肝脏修复损伤的重要因素。Objective To investigate the expression of CCL21 and CCL25 in different degree impaired liver.Methods Hepatic tissue samples were obtained from 9 patients with normal liver,10 patients with fibrotic liver,11 patients with cirrhosis.The expression of CCL21 and CCL25 was assayed by ELISA.Results The concentration of CCL21 was（0.14±0.21）pg/mg in normal liver,（0.92±0.69）pg/mg in fibrotic liver and（3.22±1.45）pg/mg in cirrhosis.The level of CCL21 expression in sclerous liver was remarkably higher than that in normal and fibrotic liver,but there was no significant difference between that in the fibrotic liver and in the normal liver.The concentration of CCL25 was（4.06±2.23）pg/mg in normal liver,（14.46±4.10）pg/mg in fibrotic liver and（21.97±2.98）pg/mg in cirrhosis.The level of CCL25 expression in sclerous liver was remarkably higher than that in normal and fibrotic liver,and the level of CCL25 in fibrotic liver was remarkably higher than that in normal liver.Conclusion With the aggravation of hepatic injury,the expression of CCL21 and CCL25 were up-regulated gradually.It suggested that there were likely to be chemokines CCL21 and CCL25 play an important role in the migrating of bone marrow derived liver stem cells to the liver participating in repair.
