免疫转印技术对人结肠Mr40KD蛋白粗提物中干扰成份的分析

摘　　要：目的；分析ＥＬＩＳＡ包被抗原在检测溃疡性结肠炎血清ＡＣＡ的非特异性干扰物。方法：应用免疫转印技术结合酶标抗人ＩｇＧ抗体和荧光抗人ＩｇＧ抗体对含Ｍｒ４０ＫＤ蛋白包被抗原的人结肠提取物进行纯度和免疫学性质的分析。结果；包被抗原粗提物中除含有Ｍｒ４０ＫＤ蛋白外，还含有过氧化物酶和人ＩｇＧ活性成分。结论；过氧化物酶和人ＩｇＧ活性成分是应用ＥＬＩＳＡ法检测ＵＣ血清ＡＣＡ－ＩｇＧ的非特异性干扰物。Objective: To analyze the nonspecific interferents in detecting ACA-IgG in sera of patients with UC by ELISA. Methods:Immunoblotting were ap plied for the analysis of human colonic extract enriched with Mr40KD protein, combined with HRP-anti-human IgG and Flurescent anti-human IgG. Result: Besides Mr40KD protein, there were also human IgG and peroxidase avtive components in the colonic crude exrtact. Conclusion: The human IgG and peroxidase active components were nonspecific interferents in detecting ACA-IgG in sera of patients with UC by ELISA.The purification of Mr40KD protein will be neces sary for the development of the ELISA.
