摘 要:目的建立葡萄酒中槲皮素和山奈酚的高效液相色谱化学发光测定法。方法基于在NaOH碱性介质中铁氰化钾可以直接氧化槲皮素和山奈酚产生化学发光原理,用高效液相色谱分离柱后化学发光法测定葡萄酒中槲皮素和山奈酚含量。结果槲皮素和山奈酚浓度分别在2.0×10-6~1.6×10-2g/m(lr=0.9990)和3.2×10-6~2.0×10-4g/m(lr=0.9998)范围内与化学发光强度呈良好的线性关系,检出限(S/N=3)分别为1.4×10-9g/ml和3.4×10-7g/ml,回收率分别为99.31%和99.06%。RSD分别为2.07%,1.29%。结论该法具有简便、快速、准确等优点,适合葡萄酒中槲皮素和山奈酚的测定。Objective To establish a new method for the determination of quercetin and kaempferol in wine by high performance liquid chromatography (HPLC) with chemiluminescence. Methods The procedure was based on the chemiluminescent enhancement by quercetin and kaempferol of the K3Fe(CN)6 system in an alkaline medium. The HPLC with post-column chemiluminescence was established for the determination of quercetin and kaempferol in wine. Results The standard curves were linear in the concentration ranges of 2.0×10^6-1.6×10^-2 g/ml (r=-0.999 0) for quercetin and 3.2×10^-6-2.0×10^-4 g/ml (r=0.999 8)for kaempfero respectively.The limits of detection were 1.4 ×10^-9 g/ml for quercetin and 3.4 ×10^-7 g/ml (S/N =3)for kaempfero respectively. The rates of recovery were 99.31% for quercetin and 99.06% for kaempferol. Conclusion The method is simple, rapid, accurate and applicable to the determination of quercetin and kaempferol in wine.
