摘 要:为了解大鼠肝再生中8种肝脏细胞的一碳代谢相关基因转录谱及其预示的代谢活动,分离大鼠再生肝的8种细胞,检测它们的一碳代谢相关基因表达变化,分析其表达模式及预示的生理活动.结果表明,肝星形细胞的甲硫氨酸转化为S-腺苷甲硫氨酸活动在肝再生启动阶段增强.胆管上皮细胞的S-腺苷甲硫氨酸转化为S-腺苷高半胱氨酸、N5-甲基四氢叶酸将甲基转移给甲硫氨酸活动在进展阶段增强.肝细胞、陷窝细胞、树突状细胞的聚谷氨酸水解、四氢叶酸与N5-甲酰基四氢叶酸相互转化在肝再生3个阶段增强.结论:大鼠肝再生与一碳代谢相关.To explore the transcription profiles of one carbon metabolism-related genes in 8 liver cell types during liver regeneration and the associated metabolism activities indicated by them,the cells are sorted out,their gene expression changes are detected,and their expression patterns and the predicted physiological activities are analyzed. The results suggest that synthesis activities of DNA methyltransferases 3A and 5-methyltetrahy-drofolate-homocysteine methyltransferase become more active at progressing phase of liver regeneration; the synthesis of methylenetetrahy drofolate dehydrogenase in hepatocyte,biliary epithelial cell,pit cell and dendritic cell,and that of folate hydrolase 1 in hepatocyte,sinusoidal endothelial cell,pit cell and dendritic cell are enhanced at three phase of liver regeneration. Conclusion:one carbon metabolism is associated with liver regeneration.
