摘  要:结合HPLC定量分析了藻毒素提取条件对提取纯培养铜绿微囊藻(Microcystis aeruginosa)胞内毒素效率的影响,优化并建立了胞内藻毒素Microcystin-LR(MCLR)提取流程:藻液离心后中加入w(甲醇)=75%萃取剂,冻融和超声破碎后离心收集上清液并进行固相萃取,分别以w(甲醇)=10%淋洗,以w(甲醇)=80%+w(TFA)=0.05%洗脱SPE柱,最后收集到较纯的微囊藻毒素MCLR.The extraction procedure of mierocystin-LR (MCLR) from intracellular Microcystis aeruginosa 905 was optimized. Alga slurry was collected through centrifugation, and then w (methanol) = 75% was added as extraction solvent. The mixed liquid was treated with freeze-thawing followed by ultrasonic for 0. 5 h, and the supernatant was collected for solid phase extraction (SPE). The optimized SPE conditions were also studied: C18 cartridge was eluted with w (methanol) =10% and then washed with w (methanol) =80% and w (TFA) =0. 05%. As a result, the MCLR with better purity was obtained.
