摘  要:目的研究MMP-9及Ki-67mRNA表达水平与颅底脑膜瘤侵袭性的关系。方法根据术前影像及术中情况将22例颅底脑膜瘤分为2组:侵袭组(肿瘤具有侵袭性,11例)和非侵袭组(肿瘤无侵袭性,11例)。通过实时荧光定量PCR检测MMP-9及Ki-67mRNA的表达水平,并采用免疫组化法验证其蛋白表达。结果非侵袭组MMP-9mRNA表达的相对值为0.12±0.07,侵袭组为1.88±0.77,差异有统计学意义(P<0.05)。非侵袭组Ki-67mRNA表达的相对值为0.07±0.02,侵袭组为0.23±0.18,差异无统计学意义(P>0.05)。直线回归分析显示:MMP-9mRNA与Ki-67mRNA表达不具有相关性。结论颅底脑膜瘤的侵袭性与MMP-9mRNA表达水平相关,与Ki-67mRNA表达水平无关。Objective To investigate the relationship between MMP-9 mRNA, K i-67 mRNA and the invasiveness of skull-base meningioma. Methods Twenty two patients with skull-base meningioma were randomly divided into two groups according to the preoperative imaging data and intraoperative observation: invasive group (tumor with invasiveness, 11 eases) and noninvasive group (tumor without invasiveness, 11 cases). The expression levels of MMP-9 and Ki-67 mRNA were detected by real time quantitative PCR and immunohistochemistry was used to verify the protein expression. Results The expression levels of MMP-9 mRNA in noninvasive group and invasive group were 0.12±0.07 and 1.88±0.77 respectively, significant differences were found between the two groups (P〈0.05). The expression level of Ki-67 mRNA in noninvasive group and invasive group were 0.07±0.02 and 0.23±0.18 respectively, no statistical significance was found between the two groups (P〉0.05). The expression level showed no significant correlation between MMP-9 and Ki-67 mRNA by linear regression analysis. Conclusions The invasiveness of skull base meningioma is of a positive correlation with MMP-9 mRNA expression, and no correlation with Ki-67 mRNA expression.
