组织芯片技术研究CD147和MMP-2在胃癌中的表达

摘　　要：目的研究CD147和基质金属蛋白酶-2(MMP-2)在胃癌组织中的表达及在肿瘤浸润和转移中的意义。方法利用组织芯片技术结合免疫组化SP法检测161例胃癌组织及40例癌旁正常组织中CD147和MMP-2的表达。结果胃癌组织和癌旁正常组织中,CD147的阳性表达率分别为81.4%(131/161)和25%(10/40),两者间差异有统计学意义(P<0.05);MMP-2阳性表达率分别为77.6%(123/161)和27.5%(11/40),两者间差异有统计学意义(P<0.05)。CD147的阳性表达与肿瘤大小、浸润深度、淋巴结转移和TNM分期显著相关(P<0.05),与患者的性别、年龄和肿瘤的分化程度无差异(P>0.05);MMP-2的阳性表达与肿瘤的浸润深度、淋巴结转移和TNM分期显著相关(P<0.05),与患者的性别、年龄、肿瘤的大小和分化程度无差异(P>0.05)。CD147和MMP-2在胃癌组织中的表达呈正相关(rs=0.597,P<0.05)。结论CD147和MMP-2的表达与胃癌的浸润和转移有关,二者具有正协同作用,并可作为判断胃癌侵袭性的有用指标。Objective To investigate the expression of CD147 and matrix metalloproteinases-2（MMP-2）in human gastric carcinoma,and to study their relationship with the invasion,metastasis and prognosis.Methods The expression of CD147 and MMP-2 proteins of 161 cases of gastric carcinoma and 40 cases of adjacent normal tissues were detected by immunohistochemistry on tissue microarrays.Results In gastric carcinoma and adjacent normal tissue the positive expression rate of CD147 was 81.4%（130/161）and 25%（10/40）respectively,there was significant difference between the two groups（P〈0.05）,and the positive expression rate of MMP-2 was 77.6%（123/161）and 27.5%（11/40）respectively,there was significant difference between the two groups（P〈0.05）.The expressions of CD147 were closely correlated with tumor size,depth of invasion,metastasis of lymph nodes and TNM stage（P〈0.05）,but no association was observed with the gender,age,pathological grades.The expression of MMP-2 was related to the depth of invasion,metastasis of lymph nodes and TNM stage（P〈0.05）,but not related to the gender,age,pathological grades and tumor size（P〈0.05）.There was a significant positive correlation between the expressions of CD147 and MMP-2 in gastric carcinoma（rs=0.597,P〉0.05）.Conclusion The expression of CDl47 and MMP-2 in gastric carcinoma is correlation with invasion and metastasis,and may be potential indexes of envaluating invasion and metastasis of gastric carcinoma.
