摘　　要：目的应用转化生长因子β诱导基因克隆3(βig-h3)真核表达载体pEGFP-C2/βig-h3转染人神经胶质瘤U87细胞,观察βig-h3对U87细胞侵袭力的影响。方法重组质粒pEGFP-C2/βig-h3用脂质体转染人神经胶质瘤U87细胞,采用明胶酶谱法和Transwell小室细胞侵袭实验检测转染后细胞基质金属蛋白酶(MMPs)表达水平和侵袭力的变化。结果与未转染的U87细胞和转染pEGFP-C2空载体的U87/vector相比较,在转染重组质粒pEG-FP-C2/βig-h3的U87/h3细胞中,βig-h3 mRNA和蛋白表达均显著增高,U87/h3细胞MMPs表达水平明显提高,细胞侵袭率显著增高(P均<0.01)。结论βig-h3可促进人神经胶质瘤U87细胞分泌MMPs,从而增强细胞的侵袭力,与神经胶质瘤的侵袭和转移密切相关。Objective To investigate effects of βig-h3 on the invasive potential of human glioma U87 cells by transfecting pEGFP-C2/βig-h3, eukaryotic expression vector of βig-h3 into U87 cells. Methods The recombinant plasmid was transfected into U87 cells with Lipofectamine2000. Gelatin-Zymography and Transwell chamber were explored to detect the production of MMPs and invasive potential of the transfected cells. Results pEGFP-C2/βig-h3 recombinant expression plasmid was successfully transfected into U87 cells, mRNA and protein expression of βig-h3 in U87/h3 ceils were obviously upregnlated compared to U87 and U87/vector cells（P 〈 0.01 ）. MMPs expression and invasive potential of U87/h3 cells were promoted significantly than those of U87 and U87/vector cells （P 〈 0.01 ）. Conclusion Overexpression of βig-h3 promotes the production of MMPs and strengthens the invasive potential of U87 cells, which indicates that βig-h3 may play essential roles in the invasive and metastatic processes of glioma.
