摘 要:以莲瓣兰杂交种子原生地共生萌发突破种皮后的胚为材料,研究氯化汞浓度、灭菌时间及超声波处理时间对莲瓣兰胚培养过程中真菌污染控制及胚成活情况的影响。结果表明,莲瓣兰杂交种子原生地共生萌发突破种皮后的胚灭菌过程中氯化汞浓度越高、灭菌时间和超声波处理时间越长,胚培养过程中真菌污染率越低,但是对胚造成的损伤程度高、胚成活率低。较适宜的灭菌方法是氯化汞浓度0.20%、灭菌时间10~15min、超声波处理时间4~6min,胚污染率控制在15.0%以内,胚成活率达到80.0%以上。The germinated Cymbidium tortisepalum embryo from original habitat was used in this paper. The effects of HgCl2 concentration, time of sterilization and time of ultrasonic treatment on the control fungal contamination in cultivating were studied. The results showed that high concentration of HgCl2, long time sterilization and ultrasonic treatment could reduce the fungal contermination. However, low survival rate of embryo was also obtained in these harmful conditions. So that the suitable approaches were established, including keeping the HgCl2 concentration to 0.20%, sterizating about 10-15 min and treating with ultrasonic for 4-6 min. In these conditions, the embryo survival and fungal contamination rate were above 80.0% and below 15.0%, respectively.
