Survivin siRNA对肺癌A549细胞吉西他滨化疗敏感性的影响

摘　　要：目的:探讨Survivin siRNA对肺癌A549细胞吉西他滨化疗敏感性的影响。方法:肺癌A549细胞分为siRNA转染组、吉西他滨组、siRNA转染+吉西他滨组和正常对照组;噻唑蓝比色法(MTT法)检测各组细胞增殖率的变化,流式细胞仪分析各组细胞的细胞周期和存活率。结果:Survivin siRNA+吉西他滨组细胞的增殖率和存活率均明显低于siRNA转染组和吉西他滨组(P<0.01);且siRNA+吉西他滨组与siRNA转染组、吉西他滨组比较,更高比率的A549细胞被阻滞于S期(P<0.01)。结论:siRNA沉默Survivin表达,增加了肺癌A549细胞对吉西他滨的化疗敏感性。Objective：To investigate the effects of Survivin siRNA on chemosensitivity of lung cancer cells A549 to gemcitabine in vitro.Methods：Lung cancer ceils A549 were divided into 4 groups：Survivin siRNA group, gemcitabine group, Survivin siRNA ＋ gemcitabine group and normal control group.The effects of Survivin siRNA and gemcitabine on cell proliferation, cell cycle and cell survival were analyzed by MTT assay and flow cytometry respectively.Results：The proliferation rate and survival rate of cells in Survivin siRNA ＋ gemcitabine group were obviously lower than that in Survivin siRNA group and gemcitabine group （P 〈 0.01）. Moreover, compared with Survivin siRNA group and gemcitabine group, more A549 cells in Survivin siRNA ＋ gemcit^bine group were blocked in S stage （P 〈 0.01）.Conelusions：Silenting the Survivin gene expression by siRNA can increase the chemosensitivity of lung cancer cells A549 to gemcitabine.
