摘 要:目的了解妊娠滋养细胞疾病分子生物学发病机制中ras基因的作用。方法应用PCR-RFLP(聚合酶链反应-限制性片段长度多态分析)检测绒毛膜细胞株JAR和BeWo、和36例标本(6例新鲜组织及30例石蜡标本)中H-ras、K-ras的第12位点点突变情况;应用免疫组织化学方法检测石蜡标本野生型P21和3种突变型P21蛋白的表达情况。结果所有被测标本均未见H-ras、K-ras第12位点的点突变。而野生型P21蛋白在妊娠滋养细胞疾病中存在着不同程度的表达,其表达强度依GTD恶性程度增加而递减。结论Ras基因并不以第12位点点突变的方式在GTD中发挥作用。野生型P21参与正常滋养细胞的增殖、分化过程;Objective To inevstigate the role of ras gene in the molecular biogenesis of Gestational Trophoblastic Disease(GTD).Methods The point mutations of H ras and K ras atcodon 12 were detected in 36 GTDs as well as in the JAR,BeWo Choriocarcinoma cell lines by a polymerase chaine reaction restriction fragment length polymorphisms (PCR RFLP);the expression of wild P 21 and three mutant P 21 in formalin fixed ,paraffin emedded tissues was examined using the immunohistohemical method.Results No mutation in K ras,H ras at condon 12 was etected in any of the cases. The wild type p 21 was expressed in the parraffin embedded GTDs,and the expression intensity was decreased according to the increase of malignancy.Conclusion The mutant ras gene at codon 12 does not play an important role in the pathogenesis of GTD.The wild type P 21 may regulate the proliferation and differentiation of the early normal trophoblastic cells,and this function is diminishing according to the increase of malignancy.
