摘 要:目的探讨多种MAGE基因在结直肠癌组织中的表达及意义。方法通过逆转录聚合酶链反应(1iT—PCR)检测MAGE-A1,MAGE—A3,MAGE—A4和MAGE-A10基因在56例结直肠癌配对癌组织、癌旁组织及四种(SW620,HCT116,HT-29,SW480)结肠癌肿瘤细胞系中的mRNA表达。统计学方法利用SPSS13.0统计软件,采用χ2检验分析。结果MAGE—A1,MAGE—A3,MAGE—A4和MAGE—A10基因在结直肠癌癌组织中的表达分别为5%(3/56),52%(29/56),63%(35/56)和34%(19/56),在癌旁组织中不表达。结肠癌细胞系SW620,HCT116和HT-29分别表达3种MAGE基因,结肠癌细胞系SW480表达2种MAGE基因。89%(50/56)和50%(28/56)的结直肠癌组织分别表达至少一种和两种MAGE基因。MAGE—A4和MAGE—A10基因在TNMⅢ+Ⅳ期结直肠癌组织中的表达明显高于Ⅰ+Ⅱ期(P=0.02和P=0.042,P值均〈0.05)。结论MAGE—A3,MAGE—A4和MAGE—A10基因有可能成为结直肠癌免疫治疗的候选靶点;MAGE—A4和MAGE—A10基因可能是结直肠癌的预后标志物。Objective To explore the expression and significance of multiple MAGE genes in colorectal carcinoma (CRC) patients. Methods The expression of MAGE-A1, MAGE-A3, MAGE-A4 and MAGE-A10 genes were studied by using the reverse transcription polymerase chain reaction (RT-PCR) in 56 CRC patients. Results MAGE-A1, MAGE-A3, MAGE-A4 and MAGE-A10 were expressed at mRNA level in5% (3/56), 52% (29/56), 63% (35/56)and34% (19/56)respectively. Colon cancer cell line SW620, HCT116, and HT-29 expressed three MAGE genes respectively and SW480 expressed two MAGE genes. In addition, 89% (50/56) and 50% (28/56) of the 56 samples expressed at least one and two MAGE genes respectively. MAGE-A4 and MAGE-A10 genes expressions are significantly higher in stage TNM Ⅲ + Ⅳ than in stage TNM Ⅰ+ Ⅱ(P 〈 0. 05). Conclusions MAGE-A3, MAGE-A4 and MAGE- A10 genes are expressed at a 'high percentage in CRC, and they are the possible potential targets of immunotherapy. MAGE-A4 and MAGE-A10 genes expressions are significantly correlated with clinical TNM stage.
