摘  要:目的研究神经轴索导向分子Sem aphorin3C(Sem a3C),Sem aphorin3F(Sem a3F)mRNA对颞叶癫痫(TLE)大鼠海马神经轴索环路重建的调控作用。方法采用侧脑室内注射红藻氨酸(KA)制作TLE大鼠模型,用N issl染色及原位杂交的方法,分别检测致痫后1d、1w、2w、3w、4w大鼠海马的齿状回(DG),CA1区、CA3区神经细胞丢失程度以及Sem a3C、Sem a3F mRNA的表达。结果KA致痫后1d始出现神经元丢失,至4w神经元丢失明显增多。KA致痫后1w,Sem a3C、Sem a3F mRNA在海马的CA1区、Sem a3F mRNA在海马的CA3区表达明显下降,持续至3w(P<0.01),4w时恢复至正常(P>0.05);Sem a3C、Sem a3F mRNA在DG的表达,Sem a3C在CA3区的表达,实验组与对照组均无明显差别(P>0.05)。结论KA致痫后海马CA1区神经元下调Sem a3C、Sem a3F mRNA的表达,CA3区神经元下调Sem a3F mRNA的表达,可能促进TLE大鼠海马神经轴索环路重建。Objective To study the guidance functions of the axon guidance molecules,semaphorin3C(Sema3C) and semaphorin3F(Sema3F) in synaptic reorganizing in hippocampus in rats of kainic acid(KA)-induced temporal lobe epilepsy(TLE) by detecting Sema3C and Sema3F mRNA expression.Methods Adult rats were made to be a model of TLE by and intraventricular injuction of KA to induce status epilepticus(SE).At 1d,1w,2w,3w and 4w after KA-induced seizures,neurons loss was detected by Nissl staining,and Sema3C and Sema3F mRNA expression was detected respectively by insituhybridization in the dentate gyrus(DG),CA1,CA3 regions of hippocampus.Results From the first day after KA-induced SE,neurons loss was detected and increased in 4w.In the first week after KA-induced SE,Sema3C and Sema3F mRNA expression decreased in the CA1 regions,Sema3F mRNA expression decreased in the CA3 regions of the hippocampus compared with the controlled rats(P〈0.01),and lasted until 3w,then increased to a normal lever in 4w(P〉0.05).Sema3C and Sema3F mRNA expression had no difference in DG,and Sema3C mRNA expression had no difference in CA3 regions compared to the controlled group(P〈0.05).Conclusions Our date suggest that neurons in CA1 regions may down regulate the Sema3C and Sema3F mRNA expression,while neurons in CA3 regions may down regulate the Sema3F mRNA expression and thereby contribute to synaptic reorganizing in seizure hippocampus.
