摘 要:目的探讨内毒素对大鼠肝星状细胞(HSC)醛固酮的分泌及核因子-kB P65(NF—kB P65)mRNA表达的影响。方法传代培养大鼠HSC-T6,不同浓度内毒素处理细胞后分组。放射免疫法检测HSC分泌醛固酮的情况,半定量RTPCR方法检测HSC表达NF-kB P65mRNA的水平。应用方差分析、t检验和Pearson直线相关分析法进行统计学分析。结果浓度为0.01、0.1、1.0和10.0mg/L内毒素组醛固酮的分泌分别为(4.95±0.35)、(5.52±0.32)、(6.04±0.60)和(5.16±0.46)ug/L,显著高于对照组的(3.65±0.51)ug/L(t=2.9745,5.8725,6.8465,3.2065;均P〈0.05)。浓度为0.01、0.1、1.0和10.0mg/L内毒素组NF-kBP65mRNA的表达分别为0.824±0.06、1.07±0.07、1.23±0.06和0.96±0.05,也明显高于对照组的0.43±0.04(t=5.4776,6.8084,7.9382,7.5136;均P〈0.01)。10.0mg/L内毒素组醛固酮的分泌和NF—kBP65mRNA的表达均较1.0mg/L内毒素组有明显降低(t=4.3865,3.7246;均P〈0.05)。HSC中醛固酮的分泌与NF-kBP65mRNA的表达存在正相关(r=0.886,P〈0.01)。结论一定浓度范围的内毒素可以上调大鼠HSC醛固酮的分泌和NF-KBP65mRNA的表达,两者存在正相关和一定的剂量效应关系,这可能是引起肝纤维化的重要因素之一。Objective To study the effects of endotoxin on aldosteron secretion and nuclear factor-kB P65 (NF-kB P65) mRNA expression in rat hepatic stellate cell (HSC). Methods Cultured rat HSC were treated with endotoxin of different concentrations. Aldosteron secretion of HSC was detected by radio-immunoassay. NF-kB P65 mRNA expression of HSC was determined by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR). The data were analyzed by variance analysis, t test and Pearson linear regression analysis. Results Aldosteron secretions of HSCgroups treated with0.01, 0.1, 1.0 and 10.0 mg/Lendotoxin [(4.95±0.35), (5.52±0.32), (6.04±0.60) and (5.16±0.46) ug/L, respectively] were all significantly higher than that in control group(3.65±0.51) ug/L (t=2. 9745, 5. 8725, 6. 8465 and 3. 2065, respectively; all P〈0.05). NF -kB P65 mRNA expressions of HSC groups treated with 0. 01, 0. 1, 1.0 and 10. 0 mg/L endotoxin (0. 82±0. 06, 1.07±0.07, 1.23±0.06 and0.96-/0.05, respectively) were also significantly higher than that in control group 0.43±0.04 (t=5.4776, 6. 8084, 7. 9382 and 7. 5136, respectively; all P〈0.01). Both aldosteron secretion and NF-kB P65 mRNA expression in HSC group treated with 10.0 mg/L endotoxin were significantly lower than those in HSC group treated with 1.0 mg/L endotoxin (t=4.3865, 3. 7246; both P〈0. 05). In these treated HSC, aldosteron secretion was positively correlated with NF-kB P65 mRNA expression (r=0.886, P〈 0.01). Conclusions Aldosteron secretion and NF-kB P65 mRNA expression in rat HSC could be up-regulated by stimulation with endotoxin, which shows a certain dose-response relationship. This may be one of the important factors of hepatic fibrosis development.
