摘  要:目的探讨阿米洛利、白藜芦醇对大鼠肝脏缺血/再灌注损伤的保护作用。方法取成年SD大鼠48只,随机分为假手术组、缺血再灌注组、不同剂量阿米洛利、白藜芦醇药物处理组。按Pringle法建立全肝缺血/再灌注模型。6h后取血,测定血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)活性和超氧化物歧化酶(SOD)、丙二醛(MDA)含量,并取肝叶制作冰冻切片,HE染色观察其组织形态。结果与假手术组相比,肝缺血/再灌注组大鼠血清中AST、ALT活性增强,SOD活性下降,MDA含量增加。白藜芦醇和阿米洛利组与缺血/再灌组相比,肝脏功能损伤较轻,AST、ALT活性下降,SOD活性增强,MDA含量下降,且有剂量依赖性。白藜芦醇和阿米洛利组病理损害较缺血再灌注组明显减轻。结论白藜芦醇与阿米洛利对肝缺血/再灌损伤有保护作用,对临床治疗缺血性肝损伤有潜在的应用价值。Objective To study the protection of amiloride and resveratrol against hepatic ischemia - reperfusion injury in rat. Methods Forty - eight adult rats were divided randomly into sham - operation group, ischemia - reperfusion group, amiloride group and resveratol group. The amiloride group and resveratol group were further divided into subgroups treated with low, middle and high doses amiloride or resveratol respectively. Hepatic isehemia was induced by Pringle's maneuver for 40 minutes, and then reperfusion was performed for 6 hours. Blood samples were collected for measurement of alanine transaminase (ALT) and aspartate transaminase ( AST). And liver tissue samples were collected for measurement of superoxide dismutase ( SOD), malondialdehyde (MDA) and morphological study. Results Compared with sham - operation group, increased activity of AST and ALT, decreased activity of SOD and increased MDA level were found in ischemia - reperfusion group. In addition, amiloride and resveratrol administrated before hepatic ischemia reperfusion could dose - dependently alleviate hepatic injury. Conclusion Resveratrol and amiloride may protect liver from ischemia - reperfusion injury and raise the prospect for the clinical treatment of hepatic ischemia injury.
