c-FLIP在胆囊癌和胆囊炎组织中的表达及其意义

摘　　要：目的研究凋亡抑制蛋白细胞型Fas相关死亡域样白介素-1β转换酶抑制蛋白(c-FLIP)在胆囊癌和胆囊炎组织中的表达差异,探讨c-FLIP在胆囊癌发生、发展中所起的作用。方法收集49例石蜡包埋的胆囊癌标本和30例慢性胆囊炎标本,用免疫组化SP法测定c-FLIP的表达,分析其表达的差异及c-FLIP表达与肿瘤分期、分化、淋巴结转移和性别的关系。结果c-FLIP在胆囊癌组织中的阳性表达率显著高于胆囊炎组织(85.71%vs26.67%,P<0.01);c-FLIP的表达与患者性别、胆囊癌的分化程度、TNM分期及有无淋巴结转移之间的关联无统计学意义(P均>0.05)。结论c-FLIP过度表达可能是细胞发生恶变、对抗T细胞的杀伤作用甚至产生免疫耐受或免疫逃逸中的重要环节,在胆囊癌的发生、发展中起重要作用。ObjectiveTo analyze the differences of expression of apoptosis inhibitor protein cellular Fas-associated death domain-like interleukin-1β converting enzyme-like inhibitory protein（c-FLIP） in tissues of gallbladder carcinoma and cholecystitis in order to probe into its significance for the initiation and development of gallbladder carcinoma.MethodsThe samples of paraffin embedding from 49 cases of gallbladder carcinoma and 30 cases of chronic cholecystitis were recruited.The expression of c-FLIP was detected by immunohistochemical method（streptavidin-perosidase）.The different expressions of c-FLIP in tissues of gallbladder carcinoma and cholecystitis were evaluated,and the relations between the expression of c-FLIP and sex,tumor stage,differentiation grade,lymph node metastasis were analyzed.ResultsThe positive expression rate of c-FLIP in gallbladder carcinoma was significantly higher than that in cholecystitis （85.71% vs 26.67%,P〈0.01）.There were all no statistical significances between the expression of c-FLIP and sex,tumor TNM stage,differentiation grade and transmission by way of the lymphatics in carcinoma of gallbladder （all P〈0.05）.ConclusionsThe high expression level of c-FLIP plays an important role in the initiation and development of gallbladder carcinoma,by which the cancer cells may resist to killing of T cells and successfully escape the supervision of human immune system.
