加味白头翁汤对溃疡性结肠炎大鼠结肠细胞因子及黏附因子的影响

摘　　要：[目的]探讨加味白头翁汤对溃疡性结肠炎(UC)大鼠结肠细胞因子及黏附分子的影响。[方法]将84只SD健康大鼠随机分为6组,采用复合法造模后,分别给予加味白头翁汤大、中、小剂量煎液、柳氮磺吡啶(SASP)混悬液及蒸馏水2 m1灌胃,连续15 d。取结肠组织,免疫组化处理并进行图文分析,记录阳性细胞吸光度平均值,并进行统计学处理。[结果]造模是成功的;各中药治疗组、SASP组治疗均有效,其中,白头翁汤大剂量组疗效最好(P<0.01),白头翁汤中剂量组与SASP组疗效相当(P>0.05)。[结论]加味白头翁汤对UC大鼠具有良好的疗效,其机制可能是调节细胞因子网络平衡,从而使免疫功能恢复正常,控制持续扩大的炎症反应。[Objective]To explore the effect of modified BaiTouWeng decoction on cytokines and adhesion molecules of rats with ulcerative colitis.[Methods]In the experiment,eighty four SD class healthy rats were randomly divided into six groups.Except the normal group,the other five groups were made through the integration of general immunity and local stimulation（DNCB and acetic acid）to create the rat model of UC.After the model was successful,the six groups were used the high,medium and low dose of modified BaiTouWeng decoction,SASP and distilled water 2 ml respectively to treat by gastric perfusion in 15 days.After the treatment was over,sarificed the rats to obtain colon tissue which was done by immunohistochemistry.Then the samples wene observed under optical microscope,which applied the automatic image analysis system and image analysis software HPIAS-2000 to scan and record absorbance values of positive cells.Selected their average obsorption of the relative content of cytokines and adhesion molecules in the biopsy.The statistical approach and the statistical results were represented as x±s.[Results]This model made with DNCB and acetic acid was successful.Each traditional Chinese medicine treatment groups and SASP group had efficacy.Among them,the best effect was the high dose group of modified BaiTouWeng decoction,and the medium dose of modified BaiTouWeng decoction and SASP group had the considerable effect.[Conclusion]The occurrence mechanism of UC is mainly related to local mucosal immune disorder,accompanied by systemic immune dysfunction.Modified BaiTouWeng decoction has better curative effect on UC,which the mechanism is to regulate the balance of cytokine network,which helps to recover normal immune function and control the continued expansion of the inflammatory response for treatment.
