上皮性卵巢癌中Maspin、p27表达的相关性及意义

摘　　要：目的:检测上皮性卵巢癌组织中Maspin mRNA、p27mRNA的表达,探讨其作用机制及临床意义。方法:采用逆转录-聚合酶链反应技术(RT-PCR)检测31例人上皮性卵巢癌、28例卵巢良性肿瘤及10例正常卵巢组织中Maspin mRNA、p27mRNA的表达,并分析它们的相关性及其与上皮性卵巢癌患者手术-病理分期的关系。结果:Maspin mRNA和p27mRNA在上皮性卵巢癌组织中分别为0.661±0.104和0.695±0.136,与卵巢良性肿瘤(0.772±0.114和0.783±0.111)及正常卵巢组织(1.108±0.191和0.998±0.135)相比,差异有统计学意义(P<0.01),两者在Ⅰ、Ⅱ期上皮性卵巢癌中的表达水平明显高于Ⅲ、Ⅳ期。在上皮性卵巢癌组织中Maspin mRNA和p27mRNA的表达呈显著正相关(r=0.806,P<0.001)。结论:Maspin mRNA的表达降低可能与上皮性卵巢癌的发生、发展和转移有关。Maspin在上皮性卵巢癌的发生、发展中的作用机制可能与p27有关。Objective： To detect the expression levels of Maspin and p27 in ovarian epithelial cancer tissues, explore the mechanism and clinical significance. Methods： The expression levels of Maspin mRNA and p27 mRNA in 31 cases with ovarian epithelial cancer, 28 cases with ovarian benign tumor and 10 normal ovarian epithelium tissues were detected by RT - PCR. The correlation between Maspin ex- pression and p27 expression and the relationship with surgical - pathological staging were analyzed. Results ： The expression levels of Maspin mRNA and p27 mRNA in ovarian epithelial cancer were （0. 66±0. 104） and （0. 695 ±0. 136）, respectively, compared with ovarian benign tumor （ 0. 772 ± 0. 114, 0. 783 ± 0. 111 ） and normal ovarian epithelium tissues （ 1.108 ± 0. 191,0. 998 ±0. 135 ）, there was significant difference （P 〈 0. 01 ） . The expression levels of Maspin mRNA and p27 mRNA in stage Ⅰ-Ⅱ were significantly higher than those in stage Ⅲ-Ⅳ. In ovarian epithelial cancer tissues, there was a positive correlation between Maspin mRNA and p27 mRNA （ T = 0. 806, P 〈 0. 001 ） . Conclusion： The low expression of Maspin may participate in the carcinogenesis, development and metastasis of ovarian epithelial cancer, and the machanism is related to p27.
