摘  要:目的研究γ-氨基丁酸(GABA)对人外周血γδT细胞的增殖、细胞周期、凋亡的变化、杀伤活性的影响及其可能的作用机制。方法在体外用不同浓度的GABA与人γδT细胞作用后,用MTT比色法和流式细胞仪(FCM)以及乳酸脱氢酶(LDH)法分别检测人γδT细胞的增殖能力、T细胞周期、凋亡和杀伤活性的变化。结果GABA能显著抑制γδT细胞的生长(P<0.01),并具有浓度依赖性;GABA显著影响细胞周期比例的分布,促进细胞凋亡;GABA抑制γδT细胞对人胰腺癌细胞株SW-1990的杀伤活性。结论GABA能显著抑制γδT细胞的增殖,抑制γδT细胞的杀伤活性。提示GABA可能是通过影响细胞周期及凋亡对γδT细胞发挥作用,进而影响其杀伤活性。Objective To research the effects of GABA on proliferation, cell cycle, apoptosis, eytotoxicity and possible mechanism of action of human peripheral blood γδT cells. Methods Used the isopentenyl pyrophosphate(IPP) method to amplify human peripheral blood γδT cells in vitro. After different doses of GABA to treat with γδT cells,MTT method was used to detect the proliferation of γδT cells; And then investigated the cell cycle and apoptosis of γδT cells by flow cytometry; Meanwhile,measured to cytotoxicity of γδT cells by lactate dehydrogenase method. Results GABA could decrease the proliferation of γδT cells in a dosedependent manner(P〈0.01). The cell cycle of γδT cells was greatly affected by treated with GABA,and the apoplosis was in creased. GABA could restraint γδT cells killing the SW-1990 cell line. Conclusion GABA may effect on γδT cells through cell cycle and apoptosis,and then affect the cytotoxicity of γδT ceils.
