摘  要:目的研究益智汤对转基因阿尔茨海默病(AD)模型小鼠脑组织超微结构的影响,探讨益智汤治疗AD的作用机制。方法APP695转基因小鼠分为AD模型组和益智汤治疗组,同月龄同背景转基因阴性小鼠为正常对照组。3个月后,使用电镜技术观察各组小鼠脑组织海马超微结构的变化。结果与对照组比较,模型组小鼠海马神经细胞出现线粒体和突触损伤,血脑屏障结构异常。益智汤组小鼠神经细胞病理变化有所减轻。结论APP695转基因小鼠脑组织超微结构出现病理变化,益智汤可通过减轻神经元线粒体和突触的损伤及改善血脑屏障的功能来维持神经细胞结构和功能的正常。Objective To observe the influence of Yizhi decoction on ultrastructural changes of hippocampus in APP695 transgenic mice with Alzheimer's disease (AD) and explore the mechanism of Yizhi decoction to AD.Methods 3-month old APP695 transgenic mice were randomly divided into model and Yizhi treatment groups,and negative transgenic mice were selected into normal control group.The ultrastructure in the hippocampal CA1 region of the mice was observed by electronic microscope after 3 months later.Results The ultrastructure in the brain of mice in transgenic group had pathological changes compared with negative group.It showed swollen,enlarged mitochondria,degenerative synaptic structure and altered blood-brain barrier structure in the hippocampal CA1 region of model group.These ultrastructural pathological changes in the Yizhi treatment group were not apparent and there were little pathological changes in the normal control group.Conclusions The ultrastructure in brain of APP695 transgenic mice has pathological changes.Yizhi decoction can protect the structure of nerve cells and blood-brain barrier,relieve its ultrastructure pathological changes,which may be its morphological basis for preventing and treating AD.
