摘 要:目的分析结直肠癌与毗邻癌旁组织中miRNA-221差异表达状况。方法常规抽提30例结直肠癌及对照癌旁组织中总RNA,应用实时荧光定量PCR方法检测标本中miRNA-221的表达状况。结果与癌旁组织相比,结直肠癌组织中miRNA-221表达明显上调(P<0.01)。结论实时荧光定量PCR是一种较精确的miRNA定量检测方法;miRNA-221可能通过转录后基因沉默机制促进结直肠癌的发生发展。Objective To investigate the different expression of miRNA-221 in colorectal carcinoma and adjacent ontumorous tissues. Methods Thirty colorcctal carcinoma and adjacent non-tumorous tissues were collected respectively and the total RNA was isolated routinely. The miRNA-221 expression was detected by using real-time quantitative polymerase chain reaction (PCR). Results The expression of miRNA-221 was significantly increased in coloreetal carcinoma tissues than those in adjacent non-tumorous tissues (P 〈0.01 ). Conclusion The real-time quantitative PCR assay is an accurate, rapid and less-expensive method to detect miRNA, miRNA-221 could promote colorectal carcinoma occurrence by means of post-transcriptional gene silence.
