摘  要:目的:研究阿尔茨海默病APPswe/PS1dE9双转基因小鼠对挥发性吸入麻醉药药物敏感性的变化。方法:C57BL/6J雌鼠与C3H/HeJ雄鼠交配得到F1代,再将F1代与3月龄的APPswe/PS1dE9双转基因小鼠交配得到F2代。F2代小鼠提取鼠尾DNA,PCR扩增目的基因并鉴定。选8月龄F2代同窝小鼠根据是否含APP和PS1基因分为转基因组和野生对照组,用夹尾法分别测定两组异氟醚、七氟醚和地氟醚3种挥发性吸入麻醉剂的最低肺泡有效浓度(MAC值)。结果:繁育F2代小鼠12只,其中转基因组7只,野生对照组5只。转基因组小鼠3种挥发性吸入麻醉剂MAC值分别为异氟醚1.129±0.160,七氟醚1.900±0.153,地氟醚5.614±0.363,均较野生对照组低,但是无统计学差异(P>0.05)。结论:APP/PS1双转基因对8月龄小鼠异氟醚、七氟醚、地氟醚3种挥发性吸入麻醉剂的MAC值无明显影响。Objective To observe the change of sensitivity to inhalation anesthetics in the APPswe/PS1AE9 double transgenic mice of Alzheimer disease (AD). Methods Female C57BL/6J mice were mated with male C3H/HeJ mice and produced first filial generation (F1), then the F1 mice were mated with 3 months old APPswe/PSldE9 double transgenic mice to produce F2 generation. DNA was extracted from the tail of F2 mice and amplified by polymerase chain reaction (PCR). One brood 8 months old F2 mice were divided into transgenic group and wild-type group based on the fact that whether the mice contained APP and PSI genes or not. Then, the minimum alveolar concentration (MAC) of 3 kinds of inhalation anesthetics: isoflurane, sevoflurane, and desflurane in the two groups were tested by clamping tail method. Results There were twelve F2 mice, of which 7 were in transgenic group and 5 were in wild-type group. The MAC of isoflurane, sevoflurane, and desflurane was 1.129 ± 0.160, 1.900 ± 0.153, and 5.614 ± 0.363 in transgenic group, which were lower than those in wild-type group, but were not significantly (P 〉 0.05). Conclusion There are no significant effect of APPswe/PS1 dE9 double transgenes on the MAC of isoflurane, sevoflurane, and desflurane in mice.
