摘  要:目的:通过观察胃癌细胞核转录因子NF-κBP65蛋白与端粒酶逆转录酶(hTERT)蛋白的表达及关系,探讨消痰散结方抑制胃癌细胞增殖的作用机制。方法:采用OB胶粘贴法建立裸鼠人胃癌MKN-45原位移植瘤模型,随机数字表法分成模型组,消痰散结低、中、高剂量组,5-Fu组,每组8只,观察消痰散结方干预对胃癌组织NF-κBP65蛋白、hTERT蛋白表达的影响及相关性。结果:①消痰散结方中、高剂量组、5-Fu组NF-κBP65蛋白的阳性表达与模型组相比,P<0.05,有统计学意义;②消痰散结方中剂量组、5-Fu组hTERT蛋白的阳性表达率与模型组比较明显降低,P<0.05,有统计学意义;消痰散结方高剂量组hTERT蛋白的阳性表达率与模型组有显著差异,P<0.01;③胃癌细胞NF-κBP65蛋白与hTERT蛋白表达呈显著正相关(r=0.929,P<0.01)。结论:胃癌MKN-45细胞中存在着活化的NF-κB及高表达的hTERT蛋白,并且存在线性相关,消痰散结方可能通过其抑制NF-κB活性,阻断了NF-κB对hTERT的活性调控,使胃癌细胞增殖受到抑制,从而阻断胃癌发展,控制胃癌复发与转移。Objective: To discover the mechanism of Xiaotan Sanjie Fang (XTF), a traditional Chinese medicinal empirical Prescription for curing gastric cancer, in inhibiting proliferation of gastric cancer cells by observing its influence on the expression of human gastric cancer NF-κBP^65 protein and hTERT protein in a nude mice in situ tumor transplantation model. Methods: Using OB glue paste method, the nude mice human gastric cancer MKN -45 in situ transplantation tumor model was established, based on which 40 mice were randomly divided into 5 groups(8 mice/every group) : the blank model group, low -dose XTF group,intermediate - dose XTF group, high - dose XTF group, and 5 - Fu group. To detect gastric cancer NF-κBP^65 protein and hTERT protein expression. Results:There was a statistical meaning by comparing the mid - , high -dose XTF groups and 5 - Fu group with the model group on NF-κBP^65 protein positive expression rate, P 〈 0. 05 ; The positive expression rate of human gastric cancer hTERT protein significantly reduce by comparing the interme- diate- dose XTF group and 5 -Fu group with the model group,P 〈 0. 05, there was a statistical meaning; There was a significant difference by comparing the high - dose XTF group on the positive expression rate of human gastric cancer hTERT protein with the model group, P 〈 O. Ol ; Gastric cancer NF-κBP^65 protein and hTERT protein expression significantly show on positive correlation ( r = 0. 929, P 〈 0. 01 ). Conclusion : Activated NF-κBP^65 protein and hTERT protein exist in the gastric cancer MKN -45 cell , and show on linear correlation, XTF can inhibit the gastric cancer cells proliferation by restraining NF-κBP^65 protein activity and blocking NF-κBP^65 protein to regulate hTERT protein activity, thus block the development of gastric cancer, control of gastric cancer recurrence and metastasis.
