Th17相关细胞因子在实验性变态反应性神经炎中的作用

摘　　要：目的观察T辅助细胞17(Th17)相关细胞因子白细胞介素17(IL-17)和白细胞介素23(IL-23)在实验性自身免疫性神经炎(EAN)模型中的表达,探讨Th17细胞在EAN的作用。方法用牛周围神经鞘磷脂(BPM)免疫Lewis大鼠,观察免疫后9、15、21d大鼠的临床表现和坐骨神经中IL-17、IL-23蛋白及mRNA的表达。结果免疫后9dEAN大鼠开始出现临床症状,15d到达疾病高峰。IL-17蛋白及mRNA在免疫后第15天表达均达到最高点,以后逐渐下降,且各组与对照组相比差异显著(P<0.05)。IL-23蛋白及mRNA在免疫后9d表达最多,随后逐渐下降,9、15d与对照组相比差异显著(P<0.05)。结论IL-17可能与EAN的疾病进程有关,IL-23可能在EAN疾病诱导阶段起一定作用。Objective To explore the roles of T helper cell 17（Th17）in experimental autoimmune neuritis（EAN） by observing expressions of Th17-interrelated cytokines IL-17 and IL-23.Methods EAN was induced in Lewis rats by immunization with bovine peripheral myelin（BPM）and complete Freund＇s adjuvant（CFA）.The clinical score of rats and pathological changes in the sciatic nerve of rats were observed.Expressions of IL-17 and IL-23 protein and mRNA in the sciatic nerve were measured by immunohistochemistry and RT-PCR technology.Results Rats developed EAN with the maximum of clinical score on day 15 post-immunization（p.i）.The peak number of IL-17-positive cells and maximum of IL-17 mRNA expression in the sciatic nerve were detected on day 15 p.i.However,the peak number of IL-23-positive cells and maximum of IL-23 mRNA expression in the sciatic nerve were detected on day 9 p.i.Conclusion IL-17 plays an important role in the development of EAN and IL-23 correlates with induction of the disease.
