摘 要:目的:研究淫羊藿苷元(IT)的制备工艺。方法:采用蜗牛酶水解淫羊藿苷制备IT,以转化率为指标,通过单因素考察pH、温度、底物的浓度、酶用量、反应时间及金属离子对转化率的影响,正交试验优化制备工艺;采用MS,1H-NMR,13C-NMR鉴定水解产物。结果:酶解反应的最适条件为温度37℃、反应介质pH6.0醋酸-醋酸钠缓冲液,底物质量浓度为10 g.L-1,酶与底物比为1∶1,反应时间48 h,Na+,Ca2+,Mg2+和Zn2+对酶解反应无显著影响,Fe2+对酶解反应有抑制作用(P<0.01);反应产物相对分子质量为368,核磁图谱证实产物为IT。结论:蜗牛酶水解淫羊藿苷制备IT反应条件温和,工艺简单可靠,适合工业化生产。Objective: To optimize the conditions for enzymolysis of Icariin by Snail Hydrolase. Methods: Take conversion as index, the effects of pH, temperature, reaction time, dosage of enzyme, concentration of Icariin and metal ion on hydrosis were studied by Single-factor designs and a L9 (34) Orthogonal design. The product was characterized through MS, 1H-NMR and 13C-NMR methods. Results: The optimum enzymolysis conversion was achieved at 37℃, pH6.0 acetic acid - sodium acetate buffer solution and 48 h. The Ca2+, Mg2+ and Zn2+ could active helicase slightly but the Fe2+ inhibite enzymolysis significantly. The production was presumed as icaritin on the basis of spectral evidences. Conclusions: Snail hydrolase can be used for preparing of IT. The condition is mildness and suitable for industrialization.
