摘  要:目的:改进快速冰冻切片技术方法。方法:采用Leica-CM1900型恒冷切片机和Feather一次性刀片对手术切除的乳腺、甲状腺、胆囊、肝脏及喉等3 000例新鲜组织标本进行常规冰冻切片取材、普通胶水包埋(冰冻)、切片、染色后封片。所用试剂的组成为:AAF混合固定液(乙醇、乙酸、中性甲醛)、苏木素染色液、醇溶性伊红、梯度乙醇及二甲苯。结果:此方法制片时间10 min,镜下组织结构、切片组织切面完整、细胞形态清晰,染色佳,术后诊断符合率为98.1%。结论:该方法简便快速,冰冻切片质量好,可为快速准确的病理诊断提供保证。Aim :To improve the technological methods of fast freezing sectioning. Methods:A total of 3 000 cases of surgical removal fresh specimens of mammary gland, thyroid, gallbladder, liver, throat and etc. were sliced and studied by German Leica-CM1900 constant cold slicing machine and Japan Feather disposable blades. The ordinary freezing sectioning procedure includes material sampling, embedding (freezing) with ordinary glue, slicing and sealing piece after staining. Reagent solution consists of AAF mixed fixative solution (acetic acid, neutral formaldehyde and alcohol) ,hematoxylin stai- ning solution, alcohol-soluble eosin, gradient ethanol and xylene. Results:By the improved method, s|ice preparation time was about 10 min, intact microscopic structure and biopsy tissue were aspect, cell morphology was clear and effectively stained. Diagnostic accuracy rate after operation reached 98.1%. Conclusion :The advantages of the improved method in- clude simple and fast preparation, improved freezing sectioning quality and accurate pathological diagnosis.
