摘 要:目的:观察硫辛酸(LA)对肾缺血/再灌注(RIR)后心肌损伤的影响。方法:30只大鼠随机分为对照组、缺血/再灌注(IR)组和IR+LA组,IR组复制大鼠RIR模型,IR+LA组再灌前28 d灌胃给予LA 250 mg/(kg.d)。观察3组大鼠心肌组织丙二醛(MDA)、髓过氧化物酶(MPO)、黄嘌呤氧化酶(XO)、超氧化物歧化酶(SOD)和血浆肌酸激酶(CK)、心肌型肌酸激酶同工酶(CK-MB)及一氧化氮(NO)的变化;测定平均动脉压(pMA)、左室收缩压(pLVS)、收缩期左室内压上升最大速率(dp/dtmax)、舒张期左室内压下降最大速率(-dp/dtmax);免疫组织化学法测定心肌组织中Caspase-3蛋白表达。结果:3组间MDA、MPO、XO、SOD、CK、CK-MB、NO、Caspase-3阳性细胞率、pMA、pLVS、dp/dtmax和-dp/dtmax比较,差异均有统计学意义(F分别为38.540、48.320、25.270、28.960、38.750、42.110、36.350、49.410、28.810、37.170、23.230和33.650,P均<0.05)。结论:LA可能通过抗炎、抗氧化、升高NO含量及抑制凋亡等途径对RIR后的心肌发挥保护作用。Aim: To investigate the effects of lipoic acid (LA) on myocardium in rats suffered from renal ischemia reperfusion. Methods:A total of 30 rats were randomly divided into 3 groups: control group, ischemia reperfusion (IR) group,and IR + LA group ( n = 10). Models of renal IR were made. Contents of malondialdehyde(MDA) , myeloperoxidase (MPO) , xanthine oxidase (XO) and superoxide dismutase (SOD) in myocardia were measured. Levels of creatine kinase (CK) , creatine kinase MB isoenzyme (CK-MB) and nitric oxide (NO) in plasma were determined. The mean arterial pressure (PMA) , left ventricular systolic pressure (PLVS) , maximal rise rate of left ventricular pressure (dp/dtm~) , and the maximal fall rate of left ventricular pressure ( - dp/dtm~~) were monitored. The change of myocardial morphology and ex- pression of Caspase-3 were observed by light microscope and immunohistochemistry method. Results :Results indicated that MDA, MPO,XO, SOD, CK, CK-MB, NO, percentage of Caspase-3 positive cells, PMA , PLVS, dp/dt and --dp/dtmax bad difference among different groups ( F was 38. 540,48. 320,25. 270,28. 960,38. 750,42. 110,36. 350,49. 410,28. 810, 37. 170,23. 230, and 33. 650, P 〈 0.05 ). Conclusion : By the way of anti-inflammatory, anti-oxidation, increasing the content of NO,and inhibiting apoptosis, LA possibly protects the myocardia suffered from renal IR.
