摘  要:目的探讨右旋柠烯对乳腺癌细胞微丝骨架机构及体外侵袭行为的影响和Fascin-1相关机制。方法不同剂量右旋柠烯作用于乳腺癌细胞MDA-MB-435,激光共聚焦技术获得F-actin细胞骨架形态morphology改变;利用Transwell小室,以Matrigel和纤维粘连蛋白构建重组基底膜,检测右旋柠烯对乳腺癌侵袭人工基底膜能力的影响;应用Western Blot法检测乳腺癌细胞Fascin-1蛋白表达。结果右旋柠烯作用后乳腺癌细胞间排列较规则,细胞基底少见长而密的微刺结构;微丝结构改变的同时对基底膜的侵袭能力下降。Western Blot结果显示,Fascin-1蛋白表达明显下调(分别为2.9±0.3、12.4±0.7和19.3±0.7),与对照组(28.6±0.5)比较差异显著(P<0.05)。结论右旋柠烯通过下调Fascin-1蛋白表达,调控乳腺癌细胞微丝结构,进而抑制体外侵袭能力。Objective To explore the effects and mechanism of d-limonene on the microfilament skeleton and invasion capability of breast cancer cell line- MDA-MB-435. Methods The laser confocal microscopy was used to get the morphology of F-actin cytoskeleton of breast cancer cell. Using Matrigel and Fibronectin to construct the artificial basal membrane, the invasion inhibition effect of d-limonene was studied in Transwell cabin. The expression level of fascin-1 protein was detected by Western-blot method. Result The breast cancer cells was well-arranged aider the treatment of d-limonene and the microspikes structure in cell basilar part was hard to found.The invasion capability of breast cancer cell were prohibited by d-limonene significantly; The expression of fascin-1 was down regulated (2.9±0.3, 12.4+0.7 and 19.3±0.7 respectively) by d-limonene compared with control groups (28.6±0.5). Conclusion D-limonene can inhibit the invasion capability of breast cancer cell MDA-MB-435. Down-regulating the expression of Fascin-1 to alter the microfilament cytoskeleton may be part of the anti-cancer mechanism of d-limonene.
