摘  要:目的:探讨温热疗法对转染胞嘧啶脱氨酶基因的基因表达有无影响。方法:脂质体法将质粒G1CEACDNa转染结肠癌LOVO细胞,G418筛选抗体克隆并扩增,给予前药5-氟胞嘧啶(5-FC)进行热化疗;以beta-actin为内参照用实时荧光定量PCR检测热化疗前后CD基因表达量有无变化。结果:实时荧光定量PCR(Real-TimeRT-PCR)检测得到热化疗前CD基因表达的ct值29.53±1.1974,beta-actinct值20.82±2.7385;热化疗后CD基因表达的ct值30.285±1.201,beta-actinct值21.225±2.237;行△△Ct法进行相对定量比较无明显差异(P>0.05,t=1.4521,ν=38)。结论:温热疗法对转染G1CEACDNa的LOVO细胞CD基因的表达无明显影响。Objective: To explore the expression of CD gene in LOVO cell after thermotherapy with 5 - FC. Method: Retroviral vector GICEACDNa was transfected to the colon cancer cell LOVO through liposome technique; The cells were selectively cultured with lmg/ml G418 until positive clones were obtained; Expression of G1CEACDNa mRNA in colon cancer cell LO- VO was analysed by Real - Time RT - PCR after thermotherapy with 5 - FC. Results: Plasmid GICEACDNa was concised by enzyme Sal 1, The expression of CD gene was detected by Real - Time RT - PCR, and the ct value of CD gene expression before thermochemotherapy was 29.53± 1.1974, beta - actinct was 20.82 ± 2. 7385; the ct value of CD gene expression after thermochemotherapy was 30.285 ± 1. 201, beta - actinct was 21. 225 ± 2. 237 ; There was no difference between the two grope calculated by AACt relative quantitation .Conclusion: No Significant variety of CD gene expression can be detected after thermochemotherapy in the experiment
