摘 要:目的观察瞬时转染信号传导与转录激活因子3(STAT3)小干扰RNA(siRNA)后对人胃癌细胞SGC-7901侵袭力的影响,探讨干扰STAT3后降低胃癌细胞侵袭力的机制。方法以人胃癌细胞系SGC-7901为研究对象,培养瞬时转染特异性siRNA的SGC-7901细胞系,通过RT-PCR、免疫细胞化学、Transwell体外侵袭实验等检测该siRNA对SGC-7901细胞STAT3及MMP-9基因表达和对细胞侵袭转移能力的影响。结果STAT3 siRNA转染SGC-7901细胞48 h后,STAT3 mRNA水平下降了73.04%,MMP-9 mRNA水平下降了57.2%;Transwell体外侵袭实验显示,STAT3 siRNA组能显著抑制SGC-7901的侵袭力(P<0.01)。结论应用siRNA技术能有效抑制SGC-7901STAT3基因的表达,进而可能通过抑制MMP-9基因的表达,降低细胞的侵袭力,为以STAT3为靶向的胃癌基因治疗提供了新的思路和方法。Objective To observe the effect of siRNA targeting signal transducers and activators of transcription 3 (STATJ) on the invasion of gastric carcinoma cell and explore the mechanism. Methods The SGC-7901 cell line was studied after SGC-7901 cells with specific anti-STAT3 siRNA was transfected. The level of STAT3 and MMP-9 mRNA in the cells was measured at real time PCR,the expression of MMP- 9 protein was detected by immunnohistochemistry and the invasion of SGC-7901 cell in vitro was measured quantitatively by the invasion test of Transwell chamber. Results Compared with two control groups the level of STAT3 mRNA reduced 73.04%, MMP-9 mRNA reduced 57.2% after 48 hours and the invasion test showed that the invasiveness was visibly restrained (P 〈 0.01 ). Conclusion Silencing STAT3 can effectively inhibit the expression of STAT3 gene, then may inhibit the expression of MMP-9 gene and degrade the invasion of gastric carcinoma,which may become a new therapy for human gastric carcinoma.
