摘  要:目的研究丹参酮ⅡA磺酸钠对高脂性脂肪肝(HFL)大鼠肝脏脂质过氧化的影响,并初步探讨其作用机制。方法用高脂饲料诱发大鼠HFL模型,8周后将建模成功的大鼠随机分为模型组、丹参酮ⅡA磺酸钠(大、小剂量组)和阳性药易善复(PPC)组,每组均给予高脂饲料及相应药物,正常对照组喂以基础饲料。连续给药6周后比较各组间肝指数、血脂、血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(ASL)活性和超氧化物歧化酶(SOD)、丙二醛(MDA)的含量;检测肝脂质、肝脏组织中SOD、MDA含量并进行肝脏病理学检查。结果丹参酮ⅡA磺酸钠可显著降低HFL模型大鼠肝指数,降低血清和肝组织中TG、TC、MDA水平,降低血清LDL-C含量和ALT、AST活性;增加血清和肝组织中SOD活性及HDL-C含量,与模型组相比差异显著(P<0.05或<0.01);并可明显减少肝细胞浆内脂滴分布的程度和炎症范围。结论丹参酮ⅡA磺酸钠可通过改善肝功能及脂质代谢紊乱,减少肝中脂质沉积,减轻肝组织损伤,对高脂性脂肪肝有一定的治疗作用。Objective To study the effects of sodium tanshinone II A sulfonate (STS) on hyperlipidemic fatty liver lipid peroxidation rats and its possible mechanism. Methods HFL model rats were induced by high fat diet for 8 weeks, and divided into four groups randomly : HFL model group, STS ( H : high dose) group, STS (L: low dose ) group and polyene phosphatidylcholine capsules (PPC) group (PPC was closen to be the control medicine). The pathological changes in liver, levels of blood-fat, ALT, AST, SOD, MDA in serum, levels of TC, TG, SOD, MDA in liver were monitored after six weeks. Results STS could decrease the level of TC,TG, MDA in plasma and hepatic index,the level of serum LDL-C content and ALT, AST activities, while increase the level of SOD activity and HDL-C content in serum and liver and also reduce distribution of lipid droplet in extent and scope in liver cells. Conclusion STS could treat the hyperlipidemic fatty liver rats induced by feeding with high fat diet through improving fat metabolism disorder, reducing levels of liver lipid, lipid peroxide products and the fatty deposited in liver, and improving the function of periphery adipose tissue.
