祛风止痉方对哮喘大鼠转录因子T-bet GATA-3及TNF-α的调节作用  被引量:5

摘  要:[目的]证实祛风止痉方对哮喘大鼠治疗作用的部分机理是通过对转录因子T-bet、GATA-3及TNF-α的调节起作用的。[方法]60只大鼠随机分组为哮喘模型组、正常对照组及给药组(高中低剂量)建立大鼠哮喘模型,并分别给药。用RT-PCR检测T-bet、GATA-3表达水平,双抗体夹心ELISA法测定TNF-α浓度。[结果]模型组大鼠外周血中转录因子GATA-3高于正常对照组及给药组,比较有统计学意义(P<0.05);低剂量组表达高于高剂量组,比较有统计学意义(P<0.05);高剂量组与正常组比较无统计学意义(P>0.05);中低剂量组比较无统计学意义(P>0.05);中低两组高于正常组,比较有统计学意义(P<0.05);中剂量组与高剂量组比较无统计学意义(P>0.05)。模型组大鼠外周血中转录因子T-bet的表达量低于正常对照组及给药组,其差异具有统计学意义(P<0.05);低剂量组低于高剂量组,比较有统计学意义(P<0.05);中高剂量高于模型组,比较有统计学意义(P<0.05)。血清中TNF-α的表达,模型组高于正常组,比较有统计学意义(P<0.05);高量组低于模型组,比较有统计学意义(P<0.05)。[结论]祛风止痉方可抑制哮喘大鼠外周血中转录因子GATA-3的表达,增加转录因子T-bet的表达,以恢复Th1/Th2平衡,同时可减低TNF-α浓度,从而降低哮喘大鼠气道高反应性,达到治疗支气管哮喘的目的。[ Objective]To study the effective mechanism of QufengZhijingFang on transcription factor T- bet, GATA - 3 and TNF - αin asthmatic rats. [ Methods ] Sixty Wister rats were randomly divided into asthmatic model group, normal control group and treatment groups ( hypsi - dosage, mid - dosage and low - dosage), they were given medicine respectiveiy. We weighted the T -bet,GATA -3"expression by RT- PCR, weighted the TNF - α' level by Enzyme - linked - Immuo - Sorbent Assay (ELISA). [ Results ] The GATA - 3 expressed in model group higher than normal control group and treatment groups, it has statistical significance (P 〈 0. 05) ; low - dosage group expressed higher than hypsi - dosage group, it has statistical significance ( P 〈 0. 05) ; hypsi - dosage group compared with normal control group, there is no statistical significance ( P 〉 0. 05 ) ; mid - dosage compared with low - dosage, there is no statistical significance (P 〉 0.05 ) ; mid - dos- age and low - dosage group higher than normal control group, it has statistical significance ( P 〈 0.05 ) ; mid - dosage group compared with hypsi - dosage group, there is no statistical significance ( P 〉 0.05 ). The T - bet expressed in model group significantly higher than normal control group and treatment groups, it has statistical significance ( P 〈 0.05 ) ; low - dosage group lower than hypsi - dosage group, it has statistical significance (P 〈 0.05) ; mid - dosage and hypsi - dosage group higher than model group, it has statistical significance ( P 〈 0.05 ). The TNF - α expressed in blood serum, model group higher than normal control group, it has statistical significance ( P 〈 0.05 ) ; hypsi - dosage group lower than model group, it has statistical significance ( P 〈 O. 05). [ Conclusion ] QufengZhijingFang can reduce GATA -3 and raise T -bet expressed in peripheral blood,so that the balance Thl/Th2 can revert,meanwhile,it can reduce TNF- α expression. So that it can a- chieve the aim
