摘 要:目的研究ZD6474对鼻咽癌细胞株CNE2放射增敏作用。方法采用MTT法测定ZD6474的半数抑制浓度(50%inhibition concentration,IC50),克隆形成实验计算细胞存活率,拟合生存曲线计算放射生物学参数,流式细胞仪检测ZD6474联合放疗的凋亡率及细胞周期分布。结果单纯用药组,CNE2细胞的存活率随ZD6474浓度的增加而降低,其IC50约为2.15μmol·L^-1。ZD6474作用CNE2细胞24h后均见到放射增敏作用,增敏比为1.535±0.134。ZD6474或照射均可增加凋亡率,减少S期细胞比例及增加G2/M期细胞比例(P〈0.01)。结论ZD6474联合照射应用可提高cNE2细胞的放射敏感性并阻碍细胞增殖、促进凋亡和干扰细胞周期分布。ZD6474有望成为EGFR高表达CNE2细胞的放射增敏剂。Objective To investigate the radiosensitizing effect of a selective small molecule EGFR tyrosine kinase inhibitor(ZD6474) on nasopharyngeal carcinoma cell line CNE2. Methods 50 percent inhibition concentration(IC50) of the ZD6474 was measured by MTT assay, cell survival rate computed by clone formation test, radiobiological parameter by fitting survival curve, and apoptosis rate and cell cycle distribution of ZD6474 combined with radiotherapy detected by flow cytometer. Results In simple medication group, the survival rate of CNE2 decreased with the increase of ZD6474 concentration and the IC50 was about 2. 15R361-3mol/ L. ZD6474 presented radiosensitizing effect after 24-hour exposure and sensitizing enhancement ratio(SER) was 1. 535±0. 134. Both ZD6474 and radiation induced cell apoptosis, reduced cell proportion at S phase and increased the proportion at G2 /M phase (P〈0.01) . Conclusion ZD6474 combined with radiation could increase the radiosensitivity of CNE2 cells, inhibit cell proliferation, promote apoptosis and alter cell cycle distribution. The ZD6474 hopefully could be a radiosensitizer for high expression CNE2 cells of EGFR.
