骨肉瘤组织中XIAP mRNA、Caspase-9 mRNA的表达变化及意义

摘　　要：目的观察X染色体连锁凋亡抑制蛋白(XIAP)mRNA和Caspase-9 mRNA在骨肉瘤中的表达变化及其与骨肉瘤临床病理指标的联系。方法采用原位杂交技术检测51例骨肉瘤组织中XIAP mRNA和Caspase-9 mRNA,并以10例骨软骨瘤(骨良性病变)组织为对照(对照组)。结果在51例骨肉瘤中XIAP mRNA和Caspase-9 mRNA的表达水平与骨肉瘤的临床病理特征无明显关系,XIAP mRNA的阳性表达率显著高于对照组(P<0.05);Caspase-9 mRNA则显著低于对照组(P<0.05);XIAP mRNA与Caspase-9 mRNA的表达呈等级负相关(r=-0.467,P<0.05)。结论XIAP、Caspase-9 mRNA可能与骨肉瘤的发生、发展有关。以XIAP和Caspase-9为靶点进行诱导骨肉瘤细胞凋亡的治疗,将可能成为临床提高保肢手术成功率及患者生活质量的新治疗思路。Objective To investigate expression and significance of X-linked inhibit apoptesis protein （XIAP）mRNA and Caspase-9 mRNA in osteosareoma. Method The XIAP mRNA and Caspase-9 mRNA of 51 osteesarcoma tissues and 10 osteochondroma tissues were detected by in situ hybridization method. Results The expression rate in osteosarcoma and osteochondroma had no obvious relationship with clinicopathology characteristics （P 〉 0.05）. The positive rate and grade of XIAP mRNA were significantly higher than that in the osteochondroma tissues （P 〈 0.05 ）, while Caspase-9 mRNA was on the contrary（ P 〈 0.05）. The expression of XIAP mRNA was positively inversely correlated with the expression of Caspase- 9 mRNA（ r = - 0. 467 ,P 〈 0. 05）. Conclusions The expression of XIAP mRNA and Caspase-9 mRNA may be closely related to the carcinogenesis and development of estcosarcoma. Detection of them may help to furtherly explore of the new therapy of osteesarcoma.
