摘 要:目的探讨TNF-α对鼻咽癌放射敏感性的影响。方法裸鼠体内移植瘤实验观察CNE-2Z、H5、S1放射后的体内增殖能力,采用HE染色观察各组裸鼠移植瘤组织坏死程度、核分裂指数、肺转移率和淋巴结转移率,免疫组织化学染色和图像分析法检测不同方法处理的S1裸鼠移植瘤组织中Caspase-3和Survivin的表达。结果鼻咽癌细胞CNE-2Z存在放射敏感性异质性,S1细胞是具有放射抵抗潜能的亚克隆株。TNF-α联合放射治疗对S1裸鼠移植瘤生长有明显抑制作用(P<0.01),TNF-α联合放射治疗组中瘤组织坏死程度、核分裂指数、肺转移率和淋巴结转移率与生理盐水联合放射治疗组比无明显差异(P>0.05)。TNF-α联合放射治疗组S1裸鼠体内移植瘤细胞核中Caspase-3表达高于单独TNF-α处理组(P<0.05);各组S1裸鼠移植瘤细胞浆中Caspase-3及Survivin的表达无明显差异(P>0.05)。结论TNF-α联合放射治疗可能通过影响胞核中Caspase-3表达抑制鼻咽癌裸鼠移植瘤生长。Objective To investigate the influence of TNF-α on radiosensitivity of nasopharyngeal carcinoma (NPC). Methods Nude mice transplantation experiment in vivo was used to observe proliferation of CNE-2Z, H5 and S1 xenografts after irradiation. HE staining was used to view the necrosis, mitotic index and the rate of pulmonary metastasis and lymph node metastasis in transplanted tumors of each group. The expression of Caspase-3 and Survivin in S1 xenografts treated with different methods was measured by immunohistochemical staining and image analysis. Results The radiosensitive heterogeneity of NPC cell CNE-2Z was identified again by nude mice radiosensitivity experiment in vivo, wherein S1 was a subclone with radio-resistant potential. Tumor growth was inhibited significantly in Sl xenografts with co-treatment of TNF-α and radiation therapy (P〈0. 01). The necrosis, mitotic index and the rate of pulmonary metastasis and lymph node metastasis were not statistically significant between the group of TNF -α co-treatment with radiation and the the group of normal sodium co-treatment with radiation (P〉0.05). The expression of nuclear Caspase-3 was higher in TNF-α and radiation treated Sl xenografts when compared to TNF-α alone (P〈0.05). The expression of cytoplasmic Caspase -3 and Survivin did not have statistical significance in each group (P〉0.05). Conclusions It is suggested that TNF-α co-treatment with radiation restrains the growth of NPC nude mice transplanted tumors via the influence on nuclear Caspase-3 expression.
