摘  要:采用生物信息学方法对GenBank中的拟南芥、玉米、龙胆、福寿草和水仙等植物β-胡萝卜素羟化酶(BCH)的核苷酸和氨基酸序列进行了比对分析,进而对其组成成分、理化性质、信号肽、亚细胞定位、疏水性/亲水性、跨膜结构域、功能结构域、基序及蛋白质二级结构等重要参数进行了预测和分析。结果表明,BCH基因全长约为1256bp,具有完整的开放阅读框,长约为943bp,编码313个氨基酸,分子量为34.82kD,理论等电点为9.18,含量最丰富的氨基酸都包含Ala(10.34%)、Leu(8.7%)和Gly(8.1%);无信号肽,定位于叶绿体中的亲水性不稳定蛋白,含有3-4个跨膜结构域,一个功能结构域,二级结构均以α-螺旋和无规则卷曲为主要构件。The nucleic acid sequence and amino acid sequence of beta-carotene hydroxylase(BCH)from Arabidopsis thaliana,Zea mays,Gentiana lutea,Adonis aestivalis,and Narcissus pseudonarcissus which were registered in GenBank,were analyzed and predicted by the tools of bioinformatics in the following aspects,mainly including the composition of nucleic acid sequence and amino acid sequence,physical and chemical characters,signal peptide,subcellular localization,hydrophobicity or hydrophilicity,transmembrane topologieal structure,functional domains,motifs search,secondary structure of protein. The results as following,the full length of BCH is about 1 256 bp,contains a complete open reading frame with length of 943 bp,encodes 313amino acids,molecular weight is 34.82 kD,theoretical isoelectric point is 9.18; the most abundant amino acids all include Ala(10.34%),Leu(8.7%) and Gly(8.1%).BCH that does not have signal peptide,and locates chloroplast,is a hydrophilic and instable protein,which features three to four transmembrane topologieal structures and a functional domain.Alpha helix and random coil are main components of all secondary structures.
