摘 要:目的检测核受体PXR和耐药相关基因在人结肠癌中表达,初步探讨其相互关系在肿瘤免疫和肿瘤耐药中意义。方法分别用RT-PCR和western blot方法检测PXR、MRP2、MRP3、ABCG2、MDR1和CYP3A4在17例结肠癌组织中的表达,并进行半定量统计分析。结果与相应正常组织相比,结肠癌组织中PXR、MRP2和MRP3 mRNA水平和蛋白水平均显著增强(P<0.05)。而结肠癌组织中ABCG2、MDR1和CYP3A4 mRNA水平与相应正常组织相比没有显著差异。在癌组织和正常组织中,MRP3 mRNA水平与PXR mRNA水平呈正相关性(P<0.05)。结论 PXR、MRP2和MRP3可能在结肠癌耐药中具有重要作用,PXR调控MRP3表达可能涉及其耐药机制。This study aimed to investigate the expression of pregnane X receptor (PXR) and muhidrug resistance genes in colon cancer, and to study its significance to multidrug resistance and immunoregulation of tumor. The mRNA and protein levels of PXR, MRP2, MRP3, ABCG2, MDR1 and CYP3A4 in colon cancer and normal tissues were determined by RT-PCR and Western blotting; intensity of interesting band was quantified using Quantity One 4.2.0 software and all data were analyzed by Paired-Samples T Test and Pearson correlation test. RT-PCR and Western blotting indicated that the both mRNA and protein levels of PXR, MRP2 and MRP3 were significantly increased (P 〈 0.05) in 17 colon cancer tissues compared with the normal colon tissues. Data analysis showed the MRP3 mRNA was positively correlated with the PXR mRNA both in colon cancer tissues (r = 0.735, P = 0.001) and in normal colon tissues (r = 0.759, P 〈 0.001). All the results implied that PXR, MRP2 and MRP3 may play an important role in human colon cancer resistance against chemotherapy, and PXR may up-regulate the expression of MRP3 in the mechanism.
