摘  要:[目的]探讨不同病理学级别人脑星形细胞瘤中Rho-GT Pases家族相关Rac1/Cdc42/PAK1信号通路的作用。[方法]应用免疫组化法检测60例不同病理级别人脑星形细胞瘤组织中Rac1、Cdc42、PAK1蛋白表达,评估其与星形细胞瘤的病理临床特征的关系。[结果]①PAK1蛋白在瘤旁脑组织表达的阳性率为100.0%,在低级别和高级别星形细胞瘤中表达的阳性率分别为100.0%和50.0%,高表达率分别为60.0%和20.0%;高级别组的阳性率和高表达率均低于瘤旁脑组织和低级别组(P<0.05),且PAK1蛋白的高表达率有从瘤旁脑组织→低级别星形细胞瘤→高级别星形细胞瘤递减的趋势。②PAK1蛋白表达的阳性率在≤40岁组显著高于>40岁组,两者差别有统计学意义(P<0.05);PAK1蛋白表达的高表达率在≤40岁组与>40岁组中的差别无统计学意义(P>0.05);③PAK1蛋白表达的阳性率和高表达率与性别、部位及肿瘤最大径无关(P>0.05);④Rac1、Cdc42蛋白均为阴性表达。[结论]PAK1在星形细胞瘤中的作用可能与Rac1/Cdc42/PAK1信号通路关系不大,而可能是通过PAK1相关其他通路起作用;PAK1在了解星形细胞瘤的发生、进展和寻找治疗新靶点方面可能有参考价值。[Purpose] To investigate effect of Rac1/Cdc42/PAK1 pathway in progression of astrocytomas. [Methods] The expressions of Racl, Cdc42 and PAK1 were examined by immmunohistochemical methods, and thein correlation with clinicopathological factors were analyzed in the 60 cases with astrocytomas. [Results] ①The positive expression rate of PAK1 protein was 100.0% in tumor adjacent brain tissue,100% in low-grade astrocytoma and 50.0% in high-grade astrocytoma. The overexpression rate of PAK1 protein was 60.0% in low-grade astrocytoma and 20.0% in high-grade astrocytoma. The positive expression rate and overexpression rate was significantly lower in the high-grade astrocytomas group than those in the low-grade astrocytoma group or in tumor adjacent brain tissue group (P〈0.05). The overexpression rate of PAK1 presented a decreasing from tumor adjacent brain tissue to low-grade astrocytoma to high-grade astrocytoma. ②The positive expression rate of PAK1 protein was significantly higher in the age ≤40 years group than that in the age〉40 years group (P〈0.05); but the overexpression rate of PAK1 protein was not correlated with age (P〈0.05). ③The positive expression rate and overexpression rate of PAK1 protein was not correlated with gender, site and the maximal diameter of tumor(P〉0.05). ④ Racl and Cdc42 protein expression was negative in all the groups. [Conclusion] The effect of PAK1 in astrocytoma might not correlate to Rac1/Cdc42/PAK1 pathway, and might relate to other pathway of PAK1. PAK1 may serve as a parameter for evaluating the carcinogenesis and progression in astrocytoma and might be a possible molecular target for treatment of the disease.
