摘  要:目的 分析人直肠癌组织miR-155和miR-200c的表达差异及其与直肠癌临床病理特征的关系.方法 选取手术切除的卣肠癌标本71例,以正常黏膜为对照,采用实时荧光定量-聚合酶链反应(qRT-PCR)对miR-155和miR-200c作定量分析.结果 相对于正常黏膜组织,miR-155和miR-200c在直肠癌组织中表达上调,差异有统计学意义(P〈0.05).miR-155的表达与患者年龄有关(P〈0.05);miR-200c的表达与患者血清癌胚抗原(CEA)水平有关(P〈0.05).不同的直肠癌性别、临床分期、浸润深度、淋巴结转移、远处转移和分化程度间miR-155、miR-200c表达的差异无统计学意义(P〉0.05).结论 miR-155和miR-200c在直肠癌组织中显著表达,可能参与直肠癌的发生过程.miR-200c对于直肠癌町能具有潜在的辅助诊断、疗效判断及预后评估意义.Objective To elucidate the miR-155 and miR-200c expression in rectal callcers,and analyze the correlation of their expression with clinicopathological features.Methods Real-time PCR was used to quantify the miR-155 and miR-200c expression in 71 pairs of rectal cancer and normal mucosa samples,and the relationship of their expression and clinicopathological features was analyzed.Results The miR-155 and miR-200c expression in rectal cancers was up-regulated compared to normal mucosa (P〈0.05 respectively).Furthermore.the miR-155 expression was related to the age of rectal cancer patients(P〈0.05),and the miR-200c expression was related to serum Careino-embryonic antigen (CEA) level(P〈0.05).There was no relationship between miR-155/miR-2130c and gender,TNM staging,local invasion,lymph metastasis,distant metastasis and cell differentiation(P〉0.05).Condusion The miR-155 and miR-200c overexpression may play acertain roleinthe development of rectal cancer.MiR-200c may represent a potential indicator in the auxiliary diagnosis,therapeutic effect and prognosis of rectal cancer.
